A rapid and efficient access to renieramycin-type alkaloids featuring a temperature-dependent stereoselective cyclization.

A flexible and practical protocol for the asymmetric synthesis of renieramycin-type antitumor alkaloids is described, in which the stereoselective Pictet-Spengler cyclization of amino ester 16 and aldehyde 15 by regulating temperature and the automatic lactamization after N-deprotection of the cyclization product are exploited to rapidly construct the common pentacyclic framework. (-)-Renieramy...

Staudinger ligation: a peptide from a thioester and azide.

[reaction: see text] The technique of native chemical ligation enables the total chemical synthesis of proteins. This method is limited, however, by an absolute requirement for a cysteine residue at the ligation juncture. Here, this restriction is overcome with a new chemical ligation method in which a phosphinobenzenethiol is used to link a thioester and azide. The product is an amide with no ...

The Suzuki–Miyaura Cross-Coupling as a Versatile Tool for Peptide Diversification and Cyclization

The (site-selective) derivatization of amino acids and peptides represents an attractive field with potential applications in the establishment of structure–activity relationships and labeling of bioactive compounds. In this respect, bioorthogonal cross-coupling reactions provide valuable means for ready access to peptide analogues with diversified structure and function. Due to the complex and...

Precursors to Rapid Elevations in Intracranial Pressure

Intracranial pressure (ICP) monitoring and management have substantially improved the outcome of patients with traumatic brain injury (TBI). However, rapid elevations in ICP remain a significant problem as they may lead to secondary brain injury and worse outcome due to cerebral ischemia. Current therapy is targeted towards treating rapid ICP elevations after they occur. Ideally, anticipatory t...
