چکیده

Abstract The use of MHC tetramers to identify antigen specific T cells has become widespread among immunologists since the late 90s. Tetramers have so ubiquitous that many companies and research facilities can ship deliver some most commonly used within a matter days. But when it comes uncommon or with antigenic peptides only recently been identified, may be quite challenge find needed tetramer. In this case, researchers are left trying produce their own tetramer having another facility do for them, which lengthy process. an effort help those researchers, we developed QuickSwitch ™MHC This loaded place-holder peptide (or exiting peptide) exchanged well-binding given haplotype. way, able make any they available in hours. More importantly, our platform means monitor how much quantify occupancy. allows user not determine if question viable tetramer, but also strong weak binding piece information invaluable future studies, particular vaccine development. In Situ Peptide-MHC-II Tetramer Staining of Antigen-Specific CD4+ T Cells in Tissues

The invention of peptide-MHC-tetramer technology to label antigen-specific T cells has led to an enhanced understanding of T lymphocyte biology. Here we describe the development of an in situ pMHC-II tetramer staining method to visualize antigen-specific CD4+ T cells in tissues. This method complements other methods developed that similarly use MHC class II reagents to stain antigen-specific CD...

MHC class II tetramers.

MHC class II tetramers have emerged as an important tool for characterization of the specificity and phenotype of CD4 T cell immune responses, useful in a large variety of disease and vaccine studies. Issues of specific T cell frequency, biodistribution, and avidity, coupled with the large genetic diversity of potential class II restriction elements, require targeted experimental design. Detection of Foreign Antigen-specific CD4+Foxp3+ Regulatory T Cells by MHC Class II Tetramer and Intracellular CD154 Staining

The unrestricted population of CD4(+)Foxp3(+) regulatory T (Treg) cells, which have been known to control the expression of autoimmune diseases and protective immunity to inflammatory reactions, has led to greater appreciation of functional plasticity. Detecting and/or isolating Ag-specific CD4(+)Foxp3(+) Tregs at the single cell level are required to study their function and plasticity. In thi...

Efficient ex vivo analysis of CD4+ T-cell responses using combinatorial HLA class II tetramer staining

MHC tetramers are an essential tool for characterizing antigen-specific CD4+ T cells. However, their ex vivo analysis is limited by the large sample requirements. Here we demonstrate a combinatorial staining approach that allows simultaneous characterization of multiple specificities to address this challenge. As proof of principle, we analyse CD4+ T-cell responses to the seasonal influenza vac...
