Quantitative monitoring of galactolipid hydrolysis by pancreatic lipase-related protein 2 using thin layer chromatography and thymol-sulfuric acid derivatization

Galactolipids are the most abundant lipids on earth where they mainly found in photosynthetic membranes of plant, algae, and cyanobacteria. Pancreatic lipase-related protein 2 (PLRP2) is an enzyme with galactolipase activity allowing mammals, especially herbivores, to digest this important source fatty acids. We present a method for quantitative analysis galactolipids galactosylated products resulting from their digestion by guinea pig PLRP2 (GPLRP2), using thin-layer-chromatography (TLC), thymol-sulfuric acid as derivatization reagent scanning densitometry detection. Thymol-sulfuric has been used colorimetric detection carbohydrates. It shown here that galactosyl group not affected bound acyl glycerol, chains length number galactose residues polar head. This allowed quantifying simultaneously initial substrate all generated upon hydrolysis monogalactosyl di-octanoylglycerol (C8-MGDG) GPLRP2 single calibration C8-MGDG reference standard. The reaction products, monooctanoyl glycerol (C8-MGMG) (MGG), were identified quantified, MGG being recovered aqueous phase analyzed separate TLC analysis. therefore suitable quantify release both acids MGDG thereby shows can contribute complete further intestinal absorption
