Quantitative MALDI mass spectrometry imaging for exploring cutaneous drug delivery of tofacitinib in human skin

In skin penetration studies, HPLC-MS/MS analysis on extracts of heat-separated epidermis and dermis provides an estimate the amount drug penetrated. this study, MALDI-MSI enabled qualitative distribution endogenous molecules molecule, tofacitinib quantitative in epidermis. The delivery to was investigated a Franz diffusion cell using three different formulations (two oil-in-water creams, C1 C2 aqueous gel). Further, vitro release testing (IVRT) performed resulted fastest from gel lowest C2. ex vivo permeation showed largest retention tofacitinib, whereas, lower higher were observed for that content treated samples comparable analysis, with below LOQ. study demonstrates can be used determination epidermis, as well as, provide valuable information tofacitinib.
