PSXIV-5 Evaluation of Heat Resistance in Beef and Dairy Calves Using Peripheral Blood Mononuclear Cells and Hair Follicles Based on Heat Shock Protein 70 Genetic Polymorphisms

Abstract Heat resistance depends on the breed of cattle and is important for genetic improvement under heat stress (HS). Study objectives were to investigate using peripheral blood mononuclear cells (PBMCs) hair follicles in beef dairy calves based shock protein (HSP) 70 polymorphisms. The samples from sixty (6-month-old, 30 Hanwoo calves, Holstein calves) collected DNA extraction. HSP70 genotype was analyzed polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) before experiment. In study 1, PBMCs isolated 20 (10 calves; H-CC, H-C/-type, 10 D-CC, D-C/-type) exposed (37°C; Con 42°C; HS) 3 h. Cells harvested used mRNA gene expression via RT-qPCR. Cell proliferation measured an ELISA kit. 2, (six times every days; low-HS, medium-HS, high-HS) measured.
