چکیده

Abstract A proper balance of pro and anti-inflammatory cytokines at the fetal-maternal interface is crucial for successful embryo implantation placental formation (placentation) during early gestation. However, mechanisms controlling this inflammatory profile are not well characterized. The C-X-C motif chemokine ligand 12 (CXCL12) its receptor CXCR4 expressed by fetal maternal tissues implicated in cytokine synthesis; roles axis placentation unclear. Using an vivo sheep model, we demonstrated that suppressing CXCL12-CXCR4 signaling results dysregulated production placenta. objective current study was to determine if disrupted from gestation would result altered midgestation. On day post-breeding, osmotic pumps were surgically installed 37 ewes delivered either inhibitor (AMD3100) 1X dose, (n=8), 1.5X, or 3X, (n=8) saline (n=13) into uterine lumen ipsilateral corpus luteum 14 days. days 90 135, collected, placenta components separated analyzed. Gene expression select analyzed using Real-time qPCR. 90, mRNA significantly AMD3100 treatment tumor necrosis factor (TNF), interleukin (IL-12), transforming growth beta (TGFB) Expression TGFB also compared control on 135 whereas IL-12 135. In conclusion, modulating CXCL12-induced actions a novel approach manipulating environment when most pregnancy losses occur. Increasing our understanding will hopefully reveal methods improve reproductive success health livestock.

the effect of e-64 on the developmental competence of sheep cocs during in vitro maturation

in the present study, the effect of e-64 at different concentrations (0.5, 1, 5 and 10 µm) added to (1) the ivm medium on oocyte nuclear maturation and developmental competence of ovine oocytes, and (2) to the ivc medium on embryonic development of ovine embryos were investigated.

The CXCL12/CXCR4 chemokine ligand/receptor axis in cardiovascular disease

The chemokine receptor CXCR4 and its ligand CXCL12 play an important homeostatic function by mediating the homing of progenitor cells in the bone marrow and regulating their mobilization into peripheral tissues upon injury or stress. Although the CXCL12/CXCR4 interaction has long been regarded as a monogamous relation, the identification of the pro-inflammatory chemokine macrophage migration in...

HLA Expression at the Maternal-Fetal Interface

Pregnancy in the human presents an "immunological paradox," because of the unexpected willingness of mothers to accept genetically disparate tissues. The fact that the fetus can develop unharmed for nine months shows that protective mechanisms must exist to permit its survival. The conditions that permit the genetically dissimilar human fetus to evade rejection by its mother's immune system hav...

Expression of integrins and extracellular matrix proteins at the maternal-fetal interface during tubal implantation.

Investigation of the expression pattern of integrins and their extracellular matrix (ECM) ligands in trophoblasts at the maternal-fetal interface during tubal pregnancy may aid better understanding of the adhesion and invasion of acceptable maternal endometrium by trophoblast cells at the very early stage of human gestation. In this study, spatial and temporal alterations of integrins and ECM l...

Rab11 family expression in the human placenta: Localization at the maternal-fetal interface

Rab proteins are a family of small GTPases involved in a variety of cellular processes. The Rab11 subfamily in particular directs key steps of intracellular functions involving vesicle trafficking of the endosomal recycling pathway. This Rab subfamily works through a series of effector proteins including the Rab11-FIPs (Rab11 Family-Interacting Proteins). While the Rab11 subfamily has been well...
