چکیده
In vitro culture is widely used for characterization of primary human acute myeloid leukemia (AML) cells, but even when using optimized handling and conditions the AML cells show spontaneous in apoptosis with a gradual decrease cell viability during culture. The extent this stress-induced varies between patients, high degree associated pre-culture BCL2 levels together low BAX Heat Shock Proteins 30 90. We compared global proteomic profiles ongoing patients (i.e., less extensive versus apoptosis) after 48 h identified 7902 proteins, only 276 proteins differed significantly >25% viable cells; 192 upregulated 84 downregulated peptides) Protein interaction network analysis based on these protein three networks that included 18 proteins; most were localized to endoplasmic reticulum several them are involved or altered process stress/unfolded stress response. To conclude, heterogeneous regard response cellular stress, difference regulation differences induction and/or unfolded

Mediators of endoplasmic reticulum stress-induced apoptosis.
The efficient functioning of the endoplasmic reticulum (ER) is essential for most cellular activities and survival. Conditions that interfere with ER function lead to the accumulation and aggregation of unfolded proteins. ER transmembrane receptors detect the onset of ER stress and initiate the unfolded protein response (UPR) to restore normal ER function. If the stress is prolonged, or the ada...
متن کاملERdj5 sensitizes neuroblastoma cells to endoplasmic reticulum stress-induced apoptosis.
Down-regulation of the unfolded protein response (UPR) can be therapeutically valuable in cancer treatment, and endoplasmic reticulum (ER)-resident chaperone proteins may thus be targets for developing novel chemotherapeutic strategies. ERdj5 is a novel ER chaperone that regulates the ER-associated degradation of misfolded proteins through its associations with EDEM and the ER stress sensor BiP...
متن کاملMitochondrial and endoplasmic reticulum stress pathways cooperate in zearalenone-induced apoptosis of human leukemic cells
BACKGROUND Zearalenone (ZEA) is a phytoestrogen from Fusarium species. The aims of the study was to identify mode of human leukemic cell death induced by ZEA and the mechanisms involved. METHODS Cell cytotoxicity of ZEA on human leukemic HL-60, U937 and peripheral blood mononuclear cells (PBMCs) was performed by using 3-(4,5-dimethyl)-2,5-diphenyl tetrazolium bromide (MTT) assay. Reactive oxy...
متن کاملInvolvement of Endoplasmic Reticulum Stress in Capsaicin-Induced Apoptosis of Human Pancreatic Cancer Cells
Capsaicin, main pungent ingredient of hot chilli peppers, has been shown to have anticarcinogenic effect on various cancer cells through multiple mechanisms. In this study, we investigated the apoptotic effect of capsaicin on human pancreatic cancer cells in both in vitro and in vivo systems, as well as the possible mechanisms involved. In vitro, treatment of both the pancreatic cancer cells (P...
متن کاملDownregulation of Smac attenuates H2O2-induced apoptosis via endoplasmic reticulum stress in human lens epithelial cells
BACKGROUND Second mitochondria-derived activator of caspases (Smac) is reported to promote apoptosis. Given the important role of apoptosis in cataract development, the aim of this study was to investigate whether Smac induces human lens epithelial cell (HLEC) apoptosis via endoplasmic reticulum stress (ERS). METHODS Smac expression was examined by immunohistochemistry in anterior lens capsul...
