Protein Phosphatases: A Neglected Target Family for Drug Discovery

نویسندگان

چکیده

The gene family of protein phosphatases is a rich but under-exploited source therapeutically validated drug targets modulating signal transduction pathways. Unlike the kinase family, research and development activities have not yet yielded any approved small-molecule drugs against phosphatase. Approximately 20 years ago, phosphatase was classified as undruggable intractable. This primarily due to spectacular failure cumulated industry-wide discovery efforts develop PTP1B inhibitors. Recently, allosteric inhibitors SHP2, member entered clinical trails, which has reawakened industry’s interest towards this neglected enzyme family. contribution reviews recent R&D trends around modulators over last years, rather than providing an exhaustive review field

Drug Discovery for Neglected Diseases: Molecular Target-Based and Phenotypic Approaches

Drug discovery for neglected tropical diseases is carried out using both target-based and phenotypic approaches. In this paper, target-based approaches are discussed, with a particular focus on human African trypanosomiasis. Target-based drug discovery can be successful, but careful selection of targets is required. There are still very few fully validated drug targets in neglected diseases, an...

Cancer drug target discovery in protein-protein interaction networks

1. Background Protein-protein interaction networks Proteins are linear chain biomolecules that are the basis of functional networks in all organisms. They include classes that regulate other proteins (e.g. kinases), classes that regulate gene expression (e.g. transcription factors), and classes that control response to disease (e.g. immunoglobulins). Aspects of their social networks have been e...

Drug abuse among nurses: A neglected challenge

Drug dependence is a prominent issue in individuals of different professions, education levels, and socioeconomic classes. Authors of this paper, including the first author (as a qualified clinical nurse and nursing supervisor), aimed to address the challenges associated with substance abuse and the subsequent failure in interactions between healthcare personnel and patients referring to clinic...

Kinetoplast DNA: A Promising Drug Target for Treatment of Leishmaniasis

Leishmaniasis is a vector-borne zoonotic disease caused by various species of the genus Leishmania, (trypanosomatidae family) that is transmitted by phlebotomine sandflies. The disease can present in a range of clinical forms, including dermal lesions, metastasis mucocutaneous forms, and fatal visceral forms. In this non-systematic review, we aimed at introducing the role of kinetoplast DNA (kD...

Drug discovery and development for neglected diseases: the DNDi model

New models of drug discovery have been developed to overcome the lack of modern and effective drugs for neglected diseases such as human African trypanosomiasis (HAT; sleeping sickness), leishmaniasis, and Chagas disease, which have no financial viability for the pharmaceutical industry. With the purpose of combining the skills and research capacity in academia, pharmaceutical industry, and con...
