Protective Effect of Siegesbeckia orientalis on Pancreatic β-Cells under High Glucose-Induced Glucotoxicity

The glucotoxicity caused by long-term exposure of β-cells to high glucose (HG) conditions may lead the generation more reactive oxygen species (ROS), reduce activity antioxidant enzymes, cause cell damage and apoptosis, induce insulin secretion dysfunction. Siegesbeckia orientalis linne is a traditional folk herbal medicine used treat snake bites, rheumatoid arthritis, allergies, immune deficiencies. In this study, we evaluated protective effect S. ethanol extract (SOE) on death oxidative stress in RIN-m5f pancreatic stimulated two HG concentrations (50–100 mM). viability assay, SOE could significantly increase survival rate under HG-induced conditions. For induced condition, treatment effectively reduced ROS formation, increased content intracellular glutathione, up-regulated expression catalase, superoxide dismutase, glutathione peroxidase. As result, decrease glucotoxicity-mediated RIN-m5F β-cells. Moreover, had function regulating different HG-mediated It increasing low concentration normal level; while decreasing relatively level. Taken together, study suggests that has HG-stimulated glucotoxic environment.
