Prognostic Impact of TP53 Mutations and Tumor Mutational Load in Colorectal Cancer

چکیده

The DNA damage response (DDR) is critical for maintaining genome stability, and abnormal DDR—resulting from mutations in damage-sensing repair proteins—is a hallmark of cancer. Here, we aimed to investigate the predictive power DDR gene tumor mutational load (TML) survival outcomes cohort 22 rectal cancer patients who received pre-operative neoadjuvant therapy. Univariate analysis revealed that TML-high TP53 were significantly associated with worse overall (OS) retaining significance multivariate analyses. Kaplan–Meier analyses further showed was disease-free (p = 0.036) OS 0.024) results our patient cohort. A total 53 somatic identified samples eight (36%) containing genes, including ATM, ATR, CHEK2, MRE11A, RAD50, NBN, ERCC2 TP53. most frequently mutated gene, 0.023) Thus, data indicate TML have prognostic value may be important independent biomarkers management. This suggests determination receiving therapy should include consideration initial genetic status.

Mutation Analysis of TP53 Tumor Suppressor Gene in Colorectal Cancer in Patients from Iran (Kerman Province)

Objective(s) P53 is an important tumor suppressor, which is mutated in later stages of many cancers and leads to resistance to chemotherapy. The aim of this study was to reveal mutations of TP53 in colorectal cancer in Kerman province. Materials and Methods A total of Forty-three colon cancer specimens as paraffin block or fresh tissues, which passed stage IIIA, were selected. Three exons 5,...

Favorable prognostic impact in loss of TP53 and PIK3CA mutations after neoadjuvant chemotherapy in breast cancer.

We investigated the loss of somatic mutations in TP53 and PIK3CA in breast cancer tissue after neoadjuvant chemotherapy (NCT) and the clinical relevance of the observed mutation profiles. Samples were derived from three cohorts: Cohort 1 consisting of 206 patients undergoing NCT with matched pre- and postchemotherapy tumor tissues; Cohort 2 consisting of 158 additional patients undergoing NCT; ...

Is Bax/Bcl-2 Ratio Considered as a Prognostic Marker with Age and Tumor Location in Colorectal Cancer?

Background: Bax and Bcl-2 are the major members of Bcl-2 family whose play a key role in tumor progression or inhibition of intrinsic apoptotic pathway triggered by mitochondrial dysfunction. Therefore, the balance between pro- and anti-apoptotic members of this family can determine the cellular fate. Methods: In this study, the relative level of mRNA expression of Bax and Bcl-2 genes was deter...

Detection of Mutations in Exons 5 and 8 of Tumor Suppressor Tp53 Gene in Patients with Squamous Cell Carcinoma of Lung Hospitalized in Afzalipour Hospital, Kerman, Iran

Introduction: Despite improvements in the diagnosis and treatment of lung cancer in the past two decades, it has remained the most common cause of death from cancer worldwide. Among all genes that are mutated in lung cancer, TP53 located on chromosome 17P13/1 has a significant diagnostic and prognostic value. TP53 mutations have been extensively studied in lung cancer and TP53 mutational spectr...

TP53 mutation in colorectal cancer.

Approximately half of all colorectal cancers show p53 (TP53) gene mutations, with higher frequencies observed in distal colon and rectal tumors and lower frequencies in proximal tumors and those with the microsatellite instability or methylator phenotypes. Alterations to this gene appear to have little or no prognostic value for colorectal cancer patients treated by surgery alone, but are assoc...
