چکیده
Abstract Background: Using a randomized, double blind, placebo-controlled, crossover protocol, we have shown previously that progesterone may prevent sleep disturbances but has no action on undisturbed sleep. In study, 8 healthy postmenopausal women took daily at 2300 h for 3 wk capsule of either 300 mg or placebo. Sleep was polygraphically recorded during the last two nights and, second night, blood samples were collected 15-min intervals using an iv catheter,. During first normal under placebo and had effect. sampling procedure associated with marked disturbances, which considerably reduced progesterone: group level, mean ± SEM duration wake after onset (WASO) dropped from 152 37 min to 71 19 (P = 0.01), slow-wave (SWS) increased 53 6 79 10 0.04). Objective: To submit individual data in study new analyses designed further investigate possible mechanism(s) actions architecture. Results: Among subjects, relation could be evidenced between levels variables, progesterone-associated improvements absolute values corresponding variables By contrast, WASO SWS, significant positive correlations (Spearman test) responses (i.e. difference, night 2, value placebo) alterations caused by placebo, 1 2): WASO: rs 0.74, P 0.037, n 8; SWS: 0.86, 0.014, 7. (Pearson test yielded similar results: r 0.85, 0.008; 0.76, 0.047). Conclusions: Although they obviously need confirmed larger studies performed variety clinical conditions, present findings suggest architecture is specifically tailored restore normality rather than normality. Since SWS mainly regulated sleep-wake homeostatic mechanism relating pressure prior wakefulness, it tempting speculate might active component this mechanism.

Molecular Mechanism of Sleep-wake Regulation
Prostaglandin (PG) D2 is the most potent endogenous sleep-promoting substance thus far reported and its sleep-inducing mechanism is the best characterized at the molecular level. PGD2 is produced by lipocalin-type PGD synthase (L-PGDS) localized in the leptomeninges, choroid plexus, and oligodendrocytes in the brain and is secreted into the cerebrospinal fl uid as a sleep hormone. PGD2 stimulat...
متن کاملDiversity and Noise Effects in a Model of Homeostatic Regulation of the Sleep-Wake Cycle
Recent advances in sleep neurobiology have allowed development of physiologically based mathematical models of sleep regulation that account for the neuronal dynamics responsible for the regulation of sleep-wake cycles and allow detailed examination of the underlying mechanisms. Neuronal systems in general, and those involved in sleep regulation in particular, are noisy and heterogeneous by the...
متن کاملSTAT1 regulates the homeostatic component of visual cortical plasticity via an AMPA receptor-mediated mechanism.
Accumulating evidence points to a role for Janus kinase/signal transducers and activators of transcription (STAT) immune signaling in neuronal function; however, its role in experience-dependent plasticity is unknown. Here we show that one of its components, STAT1, negatively regulates the homeostatic component of ocular dominance plasticity in visual cortex. After brief monocular deprivation (...
متن کاملDisfacilitation and active inhibition in the neocortex during the natural sleep-wake cycle: an intracellular study.
Earlier extracellular recordings during natural sleep have shown that, during slow-wave sleep (SWS), neocortical neurons display long-lasting periods of silence, whereas they are tonically active and discharge at higher rates during waking and sleep with rapid eye movements (REMs). We analyzed the nature of long-lasting periods of neuronal silence in SWS and the changes in firing rates related ...
متن کاملEffects of bilateral lesion of the locus coeruleus on the sleep-wake cycle in the rat
Introduction: Noradrenergic cells in LC participate in the process of cortical activation and behavioral arousal. The evidence suggests that locus ceoruleus (LC) plays an important role in the sleep-wake cycle. The aim of this study was stereological estimation of cavity caused by lesion and assessment of sleep stages after bilateral lesion of the LC. Materials and Methods: Male Wistar rats we...
