چکیده

N -glycan engineering has dramatically evolved for the development and quality control of recombinant antibodies. Fc region IgG contains two -glycans whose galactose terminals on Fc-glycan have been shown to increase stability CH2 domain improve effector functions. Nicotiana benthamiana become one most attractive production systems therapeutic In this study, Varlilumab, a CD27-targeting monoclonal antibody, was transiently produced in fresh leaves soil-grown hydroponic-grown N. , resulted yield 174 618 µg/gram, respectively. However, wild-type lacked terminal residues its -glycan. Therefore, applied fine-tune antibodies plant platforms. We further co-expressed together with murine β1,4-galactosyltransferase (β1,4-GALT) modify β1,4-linked Gal residue(s) Arabidopsis thaliana β1,3-galactosylatransferase (β1,3-GALT) galactosylation. The co-expression each GALTs successfully modification structures plant-produced IgG. Notably, β1,4-GALT had 42.5% variants having (Gal) at non-reducing terminus 55.3% that plants. Concomitantly, N- glycan profile analysis β1,3-GALT demonstrated there an increased efficiency galactosylation enhancement formation Lewis structure plant-derived Taken together, our findings show first Varlilumab biantennary β1,4-galactosylated structures.

Production of complex multiantennary N-glycans in Nicotiana benthamiana plants.

In recent years, plants have been developed as an alternative expression system to mammalian hosts for the production of therapeutic proteins. Many modifications to the plant glycosylation machinery have been made to render it more human because of the importance of glycosylation for functionality, serum half-life, and the safety profile of the expressed proteins. These modifications include re...

Reduced paucimannosidic N‐glycan formation by suppression of a specific β‐hexosaminidase from Nicotiana benthamiana

Plants are attractive hosts for the production of recombinant glycoproteins for therapeutic use. Recent advances in glyco-engineering facilitate the elimination of nonmammalian-type glycosylation and introduction of missing pathways for customized N-glycan formation. However, some therapeutically relevant recombinant glycoproteins exhibit unwanted truncated (paucimannosidic) N-glycans that lack...

Engineering of human-type O-glycosylation in Nicotiana benthamiana plants

Therapeutic properties of recombinant proteins are very often affected by the composition and heterogeneity of their glycans. Conventional expression systems for recombinant pharmaceutical proteins typically do not address this problem and produce a mixture of glycoforms that are neither identical to human glycans nor optimized for enhanced efficacy. In terms of glycosylation, plants offer cert...

Nicotiana benthamiana as a Production Platform for Artemisinin Precursors

BACKGROUND Production of pharmaceuticals in plants provides an alternative for chemical synthesis, fermentation or natural sources. Nicotiana benthamiana is deployed at commercial scale for production of therapeutic proteins. Here the potential of this plant is explored for rapid production of precursors of artemisinin, a sesquiterpenoid compound that is used for malaria treatment. METHODOLOG...

N-Glycosylation of cholera toxin B subunit in Nicotiana benthamiana: impacts on host stress response, production yield and vaccine potential

Plant-based transient overexpression systems enable rapid and scalable production of subunit vaccines. Previously, we have shown that cholera toxin B subunit (CTB), an oral cholera vaccine antigen, is N-glycosylated upon expression in transgenic Nicotiana benthamiana. Here, we found that overexpression of aglycosylated CTB by agroinfiltration of a tobamoviral vector causes massive tissue necros...
