Pre-existing immunity modulates responses to mRNA boosters

چکیده

Abstract mRNA vaccines have shown high efficacy in preventing severe COVID-19, but breakthrough infections, emerging variants and waning antibody levels warranted the use of boosters. Although boosters been widely implemented, extent to which pre-existing immunity influences remains unclear. In a cohort individuals primed with mRNA-1273 or BNT162b2 vaccines, we observed that lower spike-specific before boost were associated higher fold-increase after boost, suggesting antibodies SARS-CoV-2 downmodulate immunogenicity mRNA-based vaccines. Similar effects mice received HIV-1 Our subsequent studies show against antigen encoded by vaccine itself accelerate clearance following booster vaccination, via Fc-dependent mechanisms, limiting amount available prime B cell responses. These data suggest transient downmodulation effector functions may improve other diseases. Start-up chart string, Northwestern University National Institute on Drug Abuse (NIDA, DP2DA051912)

Pre-existing immunity against Ad vectors

Pre-existing immunity against human adenovirus (HAd) serotype 5 derived vector in the human population is widespread, thus hampering its clinical use. Various components of the immune system, including neutralizing antibodies (nAbs), Ad specific T cells and type I IFN activated NK cells, contribute to dampening the efficacy of Ad vectors in individuals with pre-existing Ad immunity. In order to...

STABILIZED1 Modulates Pre-mRNA Splicing for Thermotolerance.

High-temperature stress often leads to differential RNA splicing, thus accumulating different types and/or amounts of mature mRNAs in eukaryotic cells. However, regulatory mechanisms underlying plant precursor mRNA (pre-mRNA) splicing in the environmental stress conditions remain elusive. Herein, we describe that a U5-snRNP-interacting protein homolog STABILIZED1 (STA1) has pre-mRNA splicing ac...

Pre-existing immunity against vaccine vectors – friend or foe?

Over the last century, the successful attenuation of multiple bacterial and viral pathogens has led to an effective, robust and safe form of vaccination. Recently, these vaccines have been evaluated as delivery vectors for heterologous antigens, as a means of simultaneous vaccination against two pathogens. The general consensus from published studies is that these vaccine vectors have the poten...

Pre-existing human rhinovirus infection modulates host response to secondary bacterial infections

Background Secondary bacterial infections following viral infections of the airways are well documented and are associated with increased severity of respiratory disease compared to virus or bacterial infections alone. Human rhinovirus (HRV) infections are the most common causes of exacerbations in individuals with chronic airways diseases such as asthma and COPD. Moreover, bacterial colonizati...
