چکیده
Acetylcholinesterase/Butyrylcholinesterase inhibitors are considered an effective method for treating Alzheimer’s disease (AD). In this current work, we have computationally analyzed 11 new small molecule drugs used in various neurological diseases and Donepezil, a known inhibitor of acetylcholinesterase, as positive control. We investigated these possible fundamental interactions with acetylcholinesterase butyrylcholinesterase both critical the pathophysiology disease. selected FDA approved compounds repurposing enzymes novel therapeutic option top two molecules each protein their binding energies, interactions, most commonly drug AD treatment. Cholinesterase inhibitors: new roles and therapeutic alternatives.
An important aspect of brain cholinesterase function is related to enzymatic differences. The brain of mammals contains two major forms of cholinesterases: acetylcholinesterase (AChE) and butyrylcholinesterase (BuChE). The two forms differ genetically, structurally and for their kinetics. Butyrylcholine is not a physiological substrate in mammalian brain which makes the function of BuChE of dif...
متن کاملComparative oncology approach to drug repurposing in osteosarcoma
BACKGROUND Osteosarcoma is an orphan disease for which little improvement in survival has been made since the late 1980s. New drug discovery for orphan diseases is limited by the cost and time it takes to develop new drugs. Repurposing already approved FDA-drugs can help overcome this limitation. Another limitation of cancer drug discovery is the lack of preclinical models that accurately recap...
متن کاملA Novel Multi-Modal Drug Repurposing Approach for Identification of Potent ACK1 Inhibitors
Exploiting drug polypharmacology to identify novel modes of actions for drug repurposing has gained significant attentions in the current era of weak drug pipelines. From a serendipitous to systematic or rational ways, a variety of unimodal computational approaches have been developed but the complexity of the problem clearly needs multi-modal approaches for better solutions. In this study, we ...
متن کاملDrug Repurposing from an Academic Perspective.
Academia and small business research units are poised to play an increasing role in drug discovery, with drug repurposing as one of the major areas of activity. Here we summarize project status for a number of drugs or classes of drugs: raltegravir, cyclobenzaprine, benzbromarone, mometasone furoate, astemizole, R-naproxen, ketorolac, tolfenamic acid, phenothiazines, methylergonovine maleate an...

Dementia affects 9% of the population over the age of 65.1 Most cases of dementia (65%) are due to Alzheimer’s disease (AD). Three cholinesterase inhibitors (ChEI) are currently available on the Canadian market for the treatment of AD: donepezil, rivastigmine, and galantamine. Very few environmental studies exist on the use of these medications in a nonselected population of patients with AD, a...
