Population Genetic Structure of Feral and Cultured African Catfish (Clarias gariepinus) inferred from Random Amplified Polymorphic DNA in Kano, Nigeria

Information on the genetic structure of fish is a useful means for optimizing identification stocks, stock enhancement, breeding programs, management sustainable yield and preservation diversity. The population African Catfish, Clarias gariepinus (Burchell, 1822) in cultured feral populations from Rivers Guzuguzu, Fada Magaga (Kano State) were investigated using RAPD (Random Amplified Polymorphic DNA). Using CTAB protocol, genomic DNA was extracted caudal peduncle 157 samples live specimen collected each population. Five primers used to amplify different loci by Polymerase Chain Reaction (PCR) resultant fragments analyzed agarose gel. A total 406 reproducible bands obtained four five primers. dendrogram separated C. into two distinct clades, Guzuguzu being one clade, while Cultured belonged other clade. results based RAPD-PCR profile ranged 0.012 Magaga/Cultured 0.089 Guzuguzu/Cultured. identity also between 0.885 Magaga/Guzguzu 0.998 Magaga/Cultured. Nei’s distance confirmed above information with following ranges: 0.002 Fada/Cultured 0.102 Magaga/Guzuguzu, 0.903 Magaga/Guzuguzu 1.000 respectively. In conclusion, diversity allele richness domestic comparable.
