Polymeric Nanocarriers: A Transformation in Doxorubicin Therapies

نویسندگان

چکیده

Doxorubicin, a member of the anthracycline family, is common anticancer agent often used as first line treatment for wide spectrum cancers. Doxorubicin-based chemotherapy, although effective, associated with serious side effects, such irreversible cardiotoxicity or nephrotoxicity. Those life-threatening adverse risks, responsible elongation patients’ recuperation period and increasing medical expenses, have prompted need creating novel safer drug delivery systems. Among many proposed concepts, polymeric nanocarriers are shown to be promising approach, allowing controlled selective delivery, simultaneously enhancing its activity towards cancerous cells reducing toxic effects on healthy tissues. This article chronological examination history work progress nanostructures, designed efficient doxorubicin nanocarriers, emphasis main achievements 2010–2020. Numerous publications been reviewed provide an essential summation nanopolymer types their properties, mechanisms well active targeting stimuli-responsive strategies that currently utilized in transportation field.

Trigger responsive polymeric nanocarriers for cancer therapy.

Conventional chemotherapy for the treatment of cancer has limited specificity when administered systemically and is often associated with toxicity issues. Enhanced accumulation of polymeric nanocarriers at a tumor site may be achieved by passive and active targeting. Incorporation of trigger responsiveness into these polymeric nanocarriers improves the anticancer efficacy of such systems by mod...

Nanocarriers enhance Doxorubicin uptake in drug-resistant ovarian cancer cells.

Resistance to anthracyclines and other chemotherapeutics due to P-glycoprotein (pgp)-mediated export is a frequent problem in cancer treatment. Here, we report that iron oxide-titanium dioxide core-shell nanocomposites can serve as efficient carriers for doxorubicin to overcome this common mechanism of drug resistance in cancer cells. Doxorubicin nanocarriers (DNC) increased effective drug upta...

Folate receptor targeted biodegradable polymeric doxorubicin micelles.

Biodegradable polymeric micelles, self-assembled from a di-block copolymer of poly(D,L-lactic-co-glycolic acid) (PLGA) and poly(ethylene glycol) (PEG), were prepared to achieve folate receptor targeted delivery of doxorubicin (DOX). In the di-block copolymer structure of PLGA-b-PEG, DOX was chemically conjugated to a terminal end of PLGA to produce DOX-PLGA-mPEG, and folate was separately conju...

Development of multinuclear polymeric nanoparticles as robust protein nanocarriers.

One limitation of current biodegradable polymeric nanoparticles is their inability to effectively encapsulate and sustainably release proteins while maintaining protein bioactivity. Here we report the engineering of PLGA-polycation nanoparticles with a core-shell structure that act as a robust vector for the encapsulation and delivery of proteins and peptides. The optimized nanoparticles can lo...

Polymeric nanocarriers for siRNA delivery to murine macrophages.

This work investigates the interactions of a polycationic nanocarrier with siRNA and with cells in order to better understand the capabilities and limitations of the carrier. The polycationic nanocarriers are cross-linked copolymer nanoparticles synthesized in a single-step reaction using ARGET ATRP (activators regenerated by electron transfer atom transfer radical polymerization). The polycati...
