چکیده
The main objective of the present study is to assess and improve fermentation conditions produce polyhydroxyalkanoates (PHAs), such as homopolymer poly(3-hydroxybutyrate) (PHB) copolymer poly(3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV), from renewable inexpensive industrial waste by wild type Cupriavidus necator. Untreated residual glycerol, a byproduct generated biodiesel industry, was used carbon source C. bacterial adaptation stage, glycerol concentration other medium components were adjusted enhance PHB production. Fed batch with pulse feeding strategy propionic acid solution employed for PHBV necator, previously adapted utilization, able accumulate up 4.1 g/l under optimal substrate conditions. bacterium could also synthesize 3.4 PHBV, mole fraction 3-hydroxyvalerate (3HV) 12% fed-batch pulsed feed strategy. This first report on use ideal feedstock PHA production necator strain.

Polyhydroxyalkanoates production by engineered Cupriavidus necator from waste material containing lactose.
Cupriavidus necator DSM 545 is a well-known polyhydroxyalkanoates (PHAs) producer, but unable to grow on lactose. The aim of this study was to construct a recombinant strain of C. necator that can use lactose-containing waste material such as cheese whey, to produce PHAs. One of the intracellular PHA depolymerases (phaZ1) of C. necator was chosen to insert the lacZ, lacI and lacO genes of Esche...
متن کاملQuantitative Raman Spectroscopy Analysis of Polyhydroxyalkanoates Produced by Cupriavidus necator H16
We report herein on the application of Raman spectroscopy to the rapid quantitative analysis of polyhydroxyalkanoates (PHAs), biodegradable polyesters accumulated by various bacteria. This theme was exemplified for quantitative detection of the most common member of PHAs, poly(3-hydroxybutyrate) (PHB) in Cupriavidus necator H16. We have identified the relevant spectral region (800-1800 cm-1) in...
متن کاملPoly(hydroxyalkanoate) Production by Cupriavidus necator from Fatty Waste Can Be Enhanced by phaZ1 Inactivation
Although PHAs are regarded as an effective substitute for conventional plastics for a number of medical and agricultural applications1 and food packaging2, their full-scale manufacturing is hampered by high production costs3. Factors affecting the cost of PHAs include raw materials availability, suitable process design, and downstream processing4,5. Since almost 50 % of the total production cos...
متن کاملEffect of feeding regimens on polyhydroxybutyrate production from food wastes by Cupriavidus necator.
We investigated the effects of different feeding regimens (1-pulse, stepwise, and continuous) of fermented food-waste liquid on polyhydroxybutyrate (PHB) production. The fermentation liquid was filtered with a membrane filter (pore size, 0.45 μm) to remove anaerobic microorganisms and solids and used as a carbon source for Cupriavidus necator. One-pulse feeding yielded the highest cell concentr...
متن کاملFactors Affecting Poly(3-hydroxybutyrate) Production from Oil Palm Frond Juice by Cupriavidus necator (CCUG52238T)
Factors influencing poly(3-hydroxybutyrate) P(3HB) production by Cupriavidus necator CCUG52238(T) utilizing oil palm frond (OPF) juice were clarified in this study. Effects of initial medium pH, agitation speed, and ammonium sulfate (NH(4))(2)SO(4) concentration on the production of P(3HB) were investigated in shake flasks experiments using OPF juice as the sole carbon source. The highest P(3HB...
