Plum Pox Virus Genome-Based Vector Enables the Expression of Different Heterologous Polypeptides in Nicotiana benthamiana Plants

Plant viral vectors have become a promising tool for the rapid and cost-effective production of recombinant proteins in plants. Among numerous genera viruses that been used heterologous expression, potyviruses offer several advantages, such as polyprotein expression strategy or broad host range. In our work, pAD/pAD-agro based on plum pox virus (PPV) genome were different foreign polypeptides: alfalfa mosaic capsid protein (AMV CP), zucchini yellow (ZYMV small heat-shock Cronobacter sakazakii fused with hexahistidine (sHSP-his), fragment influenza A hemagglutinin (HA2-2), PB1-F2, SARS-CoV-2 nucleocapsid (CoN2-his), its N- C-terminal fragments (CoN-1-his CoN3-his, respectively), each anchor. Particular differed their accumulation, tissue localization, stability, solubility. The accumulation rate produced polypeptides varied from low (N, fragment) to relatively high (plant CPs, N-terminal N, PB1-F2). Some preferentially accumulated roots (sHSP, fragment, PB1-F2), showing signs proteolytic degradation leaf tissues. Thus, requires an individual approach optimization. Here, we summarize several-year experiments discuss usefulness pAD/pADep vector system.
