Pilus of Streptococcus pneumoniae: structure, function and vaccine potential

نویسندگان

چکیده

The pilus is an extracellular structural part that can be detected in some Streptococcus pneumoniae ( S. ) isolates (type I pili are found approximately 30% of strains, while type II 20%). It anchored to the cell wall by LPXTG-like motifs on peptidoglycan. Two kinds have been discovered, namely, pilus-1 and pilus-2. former encoded islet 1 (PI-1) a polymer formed protein subunits RrgA, RrgB RrgC. latter 2 (PI-2) composed mainly PitB. Although not necessary for survival , they serve as basis virulence factors mediate adhesion bacteria host cells play direct role promoting adhesion, colonization pathogenesis . In addition, candidate antigens vaccines, promising potential use vaccines with combined immunization strategies. Given current understanding regarding genes, proteins, structure, biological function epidemiological relationship serotypes, immunoprotective efficacy pilins candidates we here systematically describe research status prospects provide new ideas subsequent vaccine development.

Stability and assembly of pilus subunits of Streptococcus pneumoniae.

Pili are surface-exposed virulence factors involved in bacterial adhesion to host cells. The Streptococcus pneumoniae pilus is composed of three structural proteins, RrgA, RrgB, and RrgC and three transpeptidase enzymes, sortases SrtC-1, SrtC-2, and SrtC-3. To gain insights into the mechanism of pilus formation we have exploited biochemical approaches using recombinant proteins expressed in Esc...

Structure of the competence pilus major pilin ComGC in Streptococcus pneumoniae

Type IV pili are important virulence factors on the surface of many pathogenic bacteria and have been implicated in a wide range of diverse functions, including attachment, twitching motility, biofilm formation, and horizontal gene transfer. The respiratory pathogen Streptococcus pneumoniae deploys type IV pili to take up DNA during transformation. These "competence pili" are composed of the ma...

The Streptococcus pneumoniae Pilus-1 Displays a Biphasic Expression Pattern

The Streptococcus pneumoniae pilus-1 is encoded by pilus islet 1 (PI-1), which has three clonal variants (clade I, II and III) and is present in about 30% of clinical pneumococcal isolates. In vitro and in vivo assays have demonstrated that pilus-1 is involved in attachment to epithelial cells and virulence, as well as protection in mouse models of infection. Several reports suggest that pilus-...

Streptococcus pneumoniae pilus subunits protect mice against lethal challenge.

Streptococcus pneumoniae is a major public health threat worldwide. The recent discovery that this pathogen possesses pili led us to investigate their protective abilities in a mouse model of intraperitoneal infection. Both active and passive immunization with recombinant pilus subunits afforded protection against lethal challenge with the S. pneumoniae serotype 4 strain TIGR4.

RrgA is a pilus-associated adhesin in Streptococcus pneumoniae

Adherence to host cells is important in microbial colonization of a mucosal surface, and Streptococcus pneumoniae adherence was significantly enhanced by expression of an extracellular pilus composed of three subunits, RrgA, RrgB and RrgC. We sought to determine which subunit(s) confers adherence. Bacteria deficient in RrgA are significantly less adherent than wild-type organisms, while overexp...
