Phosphorylation Impacts Cu(II) Binding by ATCUN Motifs

نویسندگان

چکیده

ATCUN (amino terminal Cu(II) and Ni(II) binding) motifs chelate ions strongly. However, the impact of phosphorylation neighboring residues on such complexation has not been elucidated. The copper(II) dissociation constants original phosphorylated peptides from human histatin-1 serum albumin were compared using spectroscopic methods. Phosphorylation markedly weakened binding. Thus, these results indicate that may be a vital mechanism governing metal ion

Inactivation of human angiotensin converting enzyme by copper peptide complexes containing ATCUN motifs.

The copper complex [KGHK-Cu]+ demonstrates catalytic inactivation of human angiotensin converting enzyme at sub-saturating concentrations, under oxidative conditions, with an observed rate constant k approximately 2.9 +/- 0.5 x 10(-2) min(-1).

Trans-regulation of RNA-binding protein motifs by microRNA

The wide array of vital functions that RNA performs is dependent on its ability to dynamically fold into different structures in response to intracellular and extracellular changes. RNA-binding proteins regulate much of this activity by targeting specific RNA structures or motifs. One of these structures, the 3-way RNA junction, is characteristically found in ribosomal RNA and results from the ...

Sequence‐specific DNA binding by AT‐hook motifs in MeCP2

MeCP2 is a chromatin-associated protein that is mutated in Rett syndrome. Its methyl-CpG-binding domain interacts with DNA containing methylated cytosine, but other modes of recruitment to the genome have also been proposed. Here, we use in vitro and in vivo assays to investigate the DNA binding specificity of two AT-hook motifs in MeCP2. One exhibits robust sequence-specific DNA binding, where...

Heparin-binding motifs and biofilm formation by Candida albicans.

Candida albicans is a leading pathogen in infections of central venous catheters, which are frequently infused with heparin. Binding of C. albicans to medically relevant concentrations of soluble and plate-bound heparin was demonstrable by confocal microscopy and enzyme-linked immunosorbent assay (ELISA). A sequence-based search identified 34 C. albicans surface proteins containing ≥1 match to ...

Different gene regulation strategies revealed by analysis of binding motifs.

Coordinated regulation of gene expression relies on transcription factors (TFs) binding to specific DNA sites. Our large-scale information-theoretical analysis of > 950 TF-binding motifs demonstrates that prokaryotes and eukaryotes use strikingly different strategies to target TFs to specific genome locations. Although bacterial TFs can recognize a specific DNA site in the genomic background, e...
