PCR-dipstick DNA Chromatography for detection of Carbapenemase Producing Carbapenem-Resistant Enterobacteriaceae

Carbapenemase producing carbapenem-resistant Enterobacteriaceae (CP-CRE) are an epidemiologically important emerging public health threat. Early and reliable detection of CP-CRE is for containment these pathogens. The aim this study was to evaluate observe the sensitivity polymerase chain reaction (PCR)-dipstick DNA chromatography (GeneFields CPE) carbapenemase genes (NDM, OXA-48, KPC, IMP) in comparison conventional PCR. In cross-sectional study, a total 145 carbapenem resistant isolates were subjected test by phenotypic mCIM genotypic- PCR PCR-dipstick Chromatography method. dipstick assay, after multiplex DNA-DNA hybridization-based system applied where amplified products can be easily interpreted observing blue line strip visually within 15 minutes without gel electrophoresis. All (100%) positive either NDM or OXA-48 whereas 142 (97.9%) detected positivity rate NDM, both among 73.8%, 51.7% 25.5% respectively. sensitivity, specificity assay 100.0%, 92.7% 98.6%, 97.2% This showed excellent agreement with CP-CRE. may robust diagnostic tool terms rapidity

To rapidly identify carbapenemase producers in Enterobacteriaceae, we developed the Carba NP test. The test uses isolated bacterial colonies and is based on in vitro hydrolysis of a carbapenem, imipenem. It was 100% sensitive and specific compared with molecular-based techniques.
