PARP inhibitors in metastatic prostate cancer: When, who, and how?

نویسندگان

چکیده

Carcinoma prostate is among the most common cancers worldwide and mainly treated in metastatic settings through androgen blockade or chemotherapy. Homologous repair deficiency fairly (germline somatic) allows targeted therapy poly ADP-ribose polymerases (PARP) inhibitors. While data backing monotherapy strong, recent evidence seems to support frontline combination as well. Genetic testing of cancer patients also needs personalization. Pre-clinical early clinical have provided insights into mechanisms management resistance This narrative review deals with optimal patient selection behind PARP inhibitor cases carcinoma prostate.

Who, when, where, and how: salvage prostate cancer with radiotherapy.

Despite the common use of postoperative radiotherapy (RT) in patients managed initially with radical prostatectomy (RP), a number of questions remain. Raldow and colleagues build their arguments around three randomized trials that indicated a significant benefit of immediate adjuvant radiotherapy in patients with high-risk features. They acknowledge that routine adjuvant radiotherapy might resu...

Colorectal Cancer Screening—Who, How, and When?

Colorectal cancer (CRC) is the third most common cancer worldwide. It is amenable to screening as it occurs in premalignant, latent, early, and curable stages. PubMed, Cochrane Database of Systematic Reviews, and national and international CRC screening guidelines were searched for CRC screening methods, populations, and timing. CRC screening can use direct or indirect tests, delivered opportun...

PARP inhibitors in breast cancer.

The therapeutic implications of DNA damage in cancer therapy have long been appreciated and form the basis of many successful cytotoxic chemotherapy and radiotherapy treatment strategies. A novel class of DNA repair defect targeted therapeutics that inhibit poly (ADP-Ribose) polymerase (PARP) are being rapidly developed in breast cancer based on exciting preliminary clinical activity as single ...

PARP Inhibitors for Cancer Therapy

Rucaparib is an inhibitor of nuclear poly (ADP-ribose) polymerases (inhibition of PARP-1 > PARP-2 > PARP-3), following a similar drug, Olaparib. It disrupts DNA repair and replication pathways (and possibly transcription), leading to selective killing of cancer cells with BRCA1/2 mutations. Rucaparib is approved for recurrent ovarian cancers with germline or somatic mutations in BRCA1/2.
