Parental Polymorphism Survey for Evaluation and Selection of Contrasting Parents for Drought Tolerance in Rice (Oryza sativa L.) by Using SSR Markers

Aim: This study aims to find the informative polymorphic Microsatellite (SSR) markers for drought tolerance between recipient parent (Pratishya) and donor (CR Dhan 801) of rice. The tolerant QTLs (qDTY1.1, qDTY2.1, qDTY3.1) from were subsequently introgressed into popular variety Pratikshya via marker-assisted backcross breeding.
 Place Duration Study: parental polymorphism was conducted at Molecular Drought Breeding Laboratory, Department Genetics Plant Central Institute Agricultural Sciences, Banaras Hindu University, Varanasi; Crop Improvement Division, ICAR-NRRI, Cuttack during Rabi 2018-19.
 Methodology: parents used (recurrent parent) CR 801 (donor parent). leaf samples collected 20-25 days old seedlings which survey after DNA extraction. genomic isolation done by using CTAB method. A set 510 SSR covering all 12 chromosomes rice in present study. standard protocol polymerase chain reaction (PCR) agarose gel electrophoresis used. fragments documented documentation system. graphical representation marker data obtained with GGT 2.0 software.
 Results: Out 510, total 108 primer pairs exhibited (Pratikshya) remaining 392 primers monomorphic. highest percentage observed on chromosome 2 (26.53%) 3 (26.46%) followed 1 (23.08%); least number 10 (14.71%). average per cent 21.28%. screening SSRs, 69 contained dinucleotide repeats, 32 showed trinucleotide repeats 5 having tetranucleotide repeats.
 Conclusion: cultivars gives basis tagging target gene, fine mapping gene chromosome, subsequent application Marker Assisted Selection (MAS) programmes. identified this be genotyping entire population introgression QTLs/genes linked grain yield reproductive stage drought.
