PacBio full length transcript sequencing and Illumina transcriptome insight into immune defense mechanism of Litopenaeus vannamei under ammonia-N stress

To investigate the immune defense mechanism of Litopenaeus vannamei (L. vannamei) exposed to high concentration environmental ammonia-N, PacBio full-length transcriptome and Illumina sequencing were performed. After ammonia-N exposure, 24,763 32,132 transcripts obtained, while 4465 3022 new genes annotated at 0 6 h after compared L. whole genome, respectively. Besides, haemocytes transcriptomes, performed 0, 48 h, obtained a total 1017 differentially expressed (DEGs), among them 160 DEGs related neuroendocrine-immune (NEI) screened, which 40 have alternative splicing form. The following experiments showed that plasma 5-HT ACh concentrations increased significantly, then returned initial level h. Moreover, cell membrane receptors (5-HT7R, ACh7R Toll3), G-protein effectors (AC2 PLC) CaM all significantly upregulated, nuclear transcription factor NF-?B was downregulated, suggesting can act on AC-PKA, PLC - PKC pathways through 5-HT7R, subsequently function together with pathway affects downstream effector proteins expression. Toll3 is also activated participate in signal transduction process simultaneously. Furthermore, expressions BGBP, PPO3, C-lectin, ALF, LYC AIF3 increased, Syntaxin Cubilin decreased. corresponding parameters including THC, PO activity, phagocytosis rate, antibacterial bacteriolytic activities All results indicate stress, 5-HT, transfer into affect finally reduce vannamei. This study contributes better exploration crustaceans under stress.
