P-394 RAS/ BRAF molecular profile in metastatic versus non-metastatic colorectal cancer

RAS and BRAF genes are crucial therapeutic prognostic markers in metastatic colorectal cancer. Once activated, these oncogenes play a key role carcinogenesis from the beginning steps until metastasis development. Several hotspot mutations were described KRAS NRAS codon 600 of gene. Herein, we interested RAS/BRAF mutation profiles cancer compared to non-metastatic tumors. We screened for byt he Idylla IVD test NRAS-BRAF, series 810 patients (CCR) which distributed as follow: 492 had lymph nodes or distant metastases at diagnosis, 318 without any metastases. screening showed no difference KRAS*distribution between (49.6%vs 49.0% respectively), but NRAS* BRAF* more frequent CCR (4.7% 4.3% vs 2.8% BRAF*in CCR). Moreover, detailed analysis revealed that G12 recurrent than non-metastasis (71% 65% respectively) while, G13D was (20% 13% cancer). Finally, noted rare G12R correlated with (3% 0.6% mCCR).The distribution higher frequency KRAS* group (52.0% 43.6%), NRAS*/BRAF* distributions. Alleles’ highlighted codons 12 13 node (respectively 73.4% 11.4% 67.6% 9% metastases), 61* associated (8.1% 3.8%). More detailed, G12A G12S alleles (12.2% 5.2% 8.1% 1.7%) while G12D G12V (43.1% 39.2% 36.2% 28.4%). However, did not notice organ site molecular profile genes. Our results KRAS, NRAS,and mutations’ ones: advent; moreover, better prognosis KRAS: gave worst prognosis.
