Optimization of in vitro micropropagation of ethnomedicinally important underexplored plant Maytenus emarginata

In vitro callus induction and shoot emergence protocols for Maytenus emarginata (Willd) Ding Hou. have been established using young leaves stem explants of in vivo growing, one half year-old plants. Plants were growing the month August 2016 at department Botany, Ch. Charan Singh University, Meerut. However, collected August, 2019. MS (Murashige Skoog) medium WPM (Woody Plant Medium), supplemented with different concentrations combinations plant growth regulators (PGRs) such as 0.5-2.0 mg/l Naphthalene acetic acid (NAA), 0.5-1.0 Benzyl Amino Purine (BAP), 2, 4-D (1.0-2.0 mg/l) 1.0-1.5 Gibberellic (GA3) temperature regimes used emergence. The presence GA3 was found necessary from both (leaf stem). best response formation leaf obtained 24°C on 0.5 NAA+1.0mg/l BAP+1.5mg/l 2.0 NAA+ BAP+ 1.0mg/l GA3. Stem exhibited degree only 0.5mg/l 1.0 BAP+1.5 Light play very crucial role because exposure or nodal section stem, to 12 hours dark period followed by PAR (photosynthetically active radiation) 27±2 °C induced after 42 days. This is first optimization report tissue culture medicinal use emarginata.
