Optimization of chitinase production from bacillus cereus smg 1.1 using response surface methodology

Shrimp shell waste are potential to be processed further into value-added products, such as N-acetylglucosamine (GlcNAc). In the production of GlcNAc, biological approach is preferred and environmentally friendly chemical treatment. Chitinase an enzyme that plays a vital role in bioprocessing shrimp GlcNAc. Previously, Bacillus cereus SMG 1.1 was isolated from fermented paste (terasi) showed ability produce chitinase. This study designed determine optimum medium for chitinase B. through Response Surface Method (RSM) using factorial design with 15 treatments. The optimization media carried out by determining factors influence Plackett-Burman followed concentration Box-Behnken design. test assess effect independent variables on activity. Placket Burman analysis shows colloidal chitin, fructose, MgSO4.5H2O were significant components affecting 1.1. developed linear model capable predicting response. highest response value achieved at 0.75% 1.5% 0.075% resulted activity 0.0016 U/ml.
