On-line two-dimensional liquid chromatography hyphenated to mass spectrometry and ion mobility-mass spectrometry for the separation of carbohydrates from lignocellulosic biomass

Lignocellulosic biomass is a promising resource of renewable energy. Its transformation to ethanol requires efficient pretreatment leading complex liquid mixtures made hundreds oxygenated analytes. A large part the released compounds belong carbohydrates family. To overcome complexity such samples, comprehensive on-line two-dimensional reversed-phase chromatography hyphenated high-resolution mass spectrometry (RPLC × RPLC-HRMS) was dedicated separation and more specifically oligomers coming from pretreated lignocellulosic biomass. The first this study consisted in optimization hyphenation (i.e. selection stationary phases, mobile sampling time, etc.). Then, analytical method applied an industrial aqueous product sulfuric acid-based wheat straw. Around 70 well-resolved chromatographic peaks corresponding were obtained. Occupation space between each dimension estimated 75%. In last study, interest ion mobility-mass addition RPLC discussed. Some examples highlighted additional that can bring mobility RPLC-IMS-HRMS method. Using four-dimensional method, analyte described by two retention times, collisional cross section molecular formula allowing reach level detail never seen for sample compositions.
