چکیده

Abstract Introduction Blood-borne cell-free nucleic acids are increasingly emerging as significant non-invasive adjuncts to current methods of disease status evaluation in cancer patients. In this study, we sought examine whether differences exist the plasma transcriptomic profile advanced melanoma patients with a high burden compared therapeutic response. Method Plasma circulating messenger ribonucleic acid (ccfmRNA) was extracted from twenty stage IV receiving immunotherapy. Pathway focused gene expression analysis performed. Patients were assessed paired blood sample collection and CT scan assessments at baseline 3 months follow up. Response Evaluation Criteria Solid Tumours (RECIST 1.1) used for tumour estimation. Result patients, CCL5, GZMB MYD88 genes significantly over-expressed (P < 0.05 versus healthy controls). or progressive disease, CCL18, CCR1, CCR4, CD274, CSF2, GBP1 response). Finally, brain metastases, EGF, GBP1, PTGS2 0.05, without metastasis). Conclusion Significant observed between various patient groups, postulate that these may be exploited identify novel targets biomarkers relevant melanoma. CCL4 CCL5 prognostic melanoma, both had higher expressions low burden. Take-home Message ccfmRNA transcripts High is favourable

Melanoma-associated antigens as messenger RNA detection markers for melanoma.

Melanoma is heterogeneous for its biological properties and melanoma-associated antigens (MAAs). This diversity is partially observed in the expression of the MAAs involved with the melanin synthesis pathway. We therefore developed a sensitive multimarker reverse transcription-PCR plus Southern blot assay using five MAAs as molecular markers to detect primary and metastatic melanoma cells. Mela...

Cell-free translation of paramyxovirus messenger RNA.

Polypeptides corresponding in electrophoretic mobility to virion polypeptides 1, 3, and 5 were made in a reticulocyte cell-free system to which 18S RNA from Sendai virus-infected cells was added. Immune precipitation was used to select relevant polypeptides from endogenous products. The cell-free product corresponding to virion polypeptide 3 (the nucleocapsid structure unit) was the most abunda...

Plasma Circulating Cell-free Nuclear and Mitochondrial DNA as Potential Blood Biomarkers for Breast Cancer

Incidence rates of breast cancer are rising and mortality rates are proportionally high in Arab countries compared to rates in developed countries. The common late diagnosis among Arab women has been related to the low participation rates of Arab women in breast cancer screening activities (Donnelly et al., 2012). In Egypt, breast cancer is still considered number one killing cancer in females ...

Circulating giant macrophages as a potential biomarker of solid tumors.

Tumor-associated macrophages (TAMs) derived from primary tumors are believed to facilitate circulating tumor cell (CTC) seeding of distant metastases, but the mechanisms of these processes are poorly understood. Although many studies have focused on the migration of CTCs, less attention has been given to TAMs that, like CTCs, derive from tumor sites. Using precision microfilters under low-flow ...

LINE-1 hypomethylation status of circulating cell-free DNA in plasma as a biomarker for colorectal cancer

Colorectal cancer (CRC) is a serious public health problem and non-invasive biomarkers improving diagnosis or therapy are strongly required. Circulating cell-free DNA (cfDNA) has been a promising target for this purpose. In this study, we evaluated the potential of long interspersed nuclear element-1 (LINE-1) hypomethylation as a blood biomarker for CRC. LINE-1 hypomethylation level in plasma c...
