چکیده
Abstract Study question Whether other forms of intercellular communication such as extracellular vesicles (EVs) participate in stem cell maintenance the human endometrium. Summary answer The present study revealed involvement EVs a novel method between endometrial mesenchymal cells (eMSC) and their surrounding niche. What is known already In endometrium, many studies have demonstrated importance mediating various physiological well pathological processes. Our group has that myometrial are candidate niche eMSC modulating biological function. We also demonstrate Notch signaling pathway involved regulation. Although classical relies on direct contact dependent interactions, this can be activated at distance by containing ligands. Therefore, we hypothesized certain ligand(s) packaged into to mediate functions. design, size, duration Sequential beading with magnetic beads coated anti-CD140b anti-CD146 antibodies was used isolate (CD140b+CD146+ cells) from tissues proliferative (n = 15) secretory 10) phase. Participants/materials, setting, methods Endometrial samples obtained women aged 41 – 52 years undergoing total abdominal hysterectomy. EMSC cocultured percentage analysed flow cytometry. Blockage secretion performed transfection Rab27a siRNA. Western blot analysis gene silencing approach validated role eMSC. therapeutic features transplanted eMSC/myometrial determined using mouse injured model. Main results chance released internalized eMSC, leading significant effect stemness Pharmacological inhibition DAPT or 1 nullified these stimulatory effects. contains high amount notch ligand JAG1, thus inducing strong activity When JAG1 silenced EVs, self-renewal clonogenic reduced. Combined transplantation improves regeneration vivo. observed feature potentially achieved elevating proliferation suppressing apoptosis Limitations, reasons for caution activities remains unknown. Gene Expression Profile of Adherent Cells Derived From Human Peripheral Blood: Evidence of Mesenchymal Stem Cells
Mesenchymal stem cells (MSCs) provide a novel option in cellular therapy and tissue engineering. Recent studies indicated that it is possible to obtain MSCs from peripheral blood by attachment ability to plastic surface. To evaluate adherent cells derived from peripheral blood, their expression profile and surface markers were investigated. The results of RT-PCR indicated that these cells expre...
متن کاملExtracellular Vesicles Derived from Osteogenically Induced Human Bone Marrow Mesenchymal Stem Cells Can Modulate Lineage Commitment
The effective osteogenic commitment of human bone marrow mesenchymal stem cells (hBMSCs) is critical for bone regenerative therapies. Extracellular vesicles (EVs) derived from hBMSCs have a regenerative potential that has been increasingly recognized. Herein, the osteoinductive potential of osteogenically induced hBMSC-EVs was examined. hBMSCs secreted negatively charged nanosized vesicles (∼ 3...
متن کاملDifferentiation Potential of Nestin (+) and Nestin (-) Cells Derived from Human Bone Marrow Mesenchymal Stem Cells into Functional Insulin Producing Cells
The feasibility of isolating and manipulating mesenchymal stem cells (MSCs) from human patients provides hope for curing numerous disease and disorders. Recent phenotypic analysis showed heterogeneity of MSCs. A nestin progenitor cell is a subpopulation within MSCs which plays a role in pancreas regeneration during embryogenesis. This study aimed to separate nestin (+) cells from human bone mar...
متن کاملDifferentiation of Mesenchymal Stem Cells Derived From Human Adipose Tissue into Cholinergic-like Cells: In Vitro Study
Introduction: Cholinergic-associated diseases currently constitute a significant cause of neurological and neurodegenerative disabilities. As the drugs are not efficient in improving the suffered tissues, stem cell treatment is considered an effective strategy for substituting the lost cells. Methods: In the current study, we set out to investigate the differentiation properties of human adip...
