O-039 Fertilization: Mechanisms of pronuclear migration and genome unification

چکیده
Abstract Most early human embryos contain aneuploid cells. Aneuploidy often arises during the mitotic divisions of embryo, but its origin remains elusive. Human zygotes that cluster their nucleoli at pronuclear interface are thought to be more likely develop into healthy euploid embryos. I will present data showing how parental genomes with in each pronucleus and bovine zygotes. Parental genome clustering is required for reliable unification after fertilization. During migration intact pronuclei, polarize toward other a process driven by centrosomes, dynein, microtubules, nuclear pore complexes. The maternal paternal chromosomes eventually interface, close proximity other, yet separated. ensures rapid upon envelope breakdown. However, fails, leading chromosome segregation errors micronuclei incompatible embryonic development. In summary, our reveal why nucleolar correlates formation addition, we used automated analysis trajectories clustering. Our results support use as parameter identify higher developmental potential.

Background: Assisted fertilization procedures are a currently widespread practice to regulate reproduction in humans and animals. The arising question is why the human being manipulating gametes to generate new individuals, if we do not understand yet the molecular mechanism of fertilization?. Successful completion of fertilization in mammals is dependent on three membrane fusion events: 1. the...

O-38: Concurrent Whole-Genome Haplotyping and Copy-Number Profiling of Single Cells
Background Methods for haplotyping and DNA copynumber typing of single cells are paramount for studying genomic heterogeneity and enabling genetic diagnosis. Before analyzing the DNA of a single cell by microarray or next-generation sequencing, a whole-genome amplification (WGA) process is required, but it substantially distorts the frequency and composition of the cell’s alleles. As a conseque...

How do pronuclei migrate towards each other? The zebrafish futile cycle gene is shown to encode a maternally expressed membrane protein required for nuclear attachment and migration along the sperm aster.
متن کاملPronuclear scoring as a predictor of embryo quality in in vitro fertilization program.
Many strategies have been proposed for the selection of viable embryos for transfer in human assisted reproduction. These have included morphological scoring criteria for 20, 28, 44 and 68 h after insemination. The embryo selection is based on morphology, degree of fragmentation and development to the 8-cell. All have shown some correlation with implantation. However, the overall success of the...
