In this review, we discuss the possibility and feasibility of nuclear factor erythroid 2-related 2 (Nrf2) as a therapeutic target to minimize devastating effects brain injury. To complete comprehensive literature searches were conducted in MEDLINE, PubMed, Embase, PsycINFO databases for English scientific peer-reviewed articles through December 2022. This short review addressed different sources oxidative stress its on blood-brain barrier (BBB) dysfunction, mitochondrial damage, changes variety inflammatory molecules associated with central nervous system (CNS) At last, explained potential efficacy Nrf2 transcription reducing stress-mediated secondary damages after CNS The role CPUY192018, an inhibitor Nrf2-Keap1 protein-protein interaction protecting injured cells is given evidence Nrf2’s activating antioxidant genes. Overall, scope developing interventions pathophysiological conditions injury-induced free radical/inflammatory signaling acknowledged. has widespread application basic clinical neuroscience understanding treating disorders, including neurological diseases. development innovative strategies using Nrf2-inducing agents can be applied reduce complications TBI before advancing it posttraumatic disorder (PTSD).
