Novel urine-based DNA methylation biomarkers for urothelial bladder carcinoma detection in patients with hematuria

چکیده

Background Urothelial bladder carcinoma (UBC) is usually detected during work-up for hematuria. Cystoscopy and/or contrast-enhanced imaging are the gold standard tools UBC diagnosis, despite limited by being invasive, expensive and low yield in small flat tumors. Objectives To assess diagnostic performance of urine-based DNA methylation six genes (GATA4, P16, P14, APC, CDH1 CD99) detection patients with hematuria.Patients methods Voided urine was collected from consecutive presented hematuria cytology assay assigned using methylation-specific Polymerase Chain Reaction (PCR). Further assessment office cystoscopy subsequent inpatient cystoscopic biopsy positive findings done. The characteristics were assessed based on its capability to predict UBC.Results We included 246 study identified macroscopic 204 (82.9%) patients. Positive found 78 (31.7%) GATA4, CD99 127 (51.6%), 52 (21.1%), 117 (47.6%), 106 (43.1%), 90 (36.6%) 71 (28.9%) patients, respectively. sensitivity ranges 35% (95%CI: 31–39) 83% 79–87). Optimal specificity (SP) (100%) noted APC genes. While other P14 CD99), SP 95% 92–98), 96% 92–99) 97% 93–99), On multivariate logistic regression analysis, all exclusively demonstrated independent prediction UBC. receiver operator characteristic (ROC) tested showed superior area under curve (AUC) when compared cytology. Conclusions have developed a novel predictive capacity over cytology.

Detection of bladder cancer using novel DNA methylation biomarkers in urine sediments.

BACKGROUND Bladder cancer (BCa) remains a lethal malignancy that can be cured if detected early. DNA hypermethylation is a common epigenetic abnormality in cancer that may serve as a marker of disease activity. METHODS We selected 10 novel candidate genes from the most frequently hypermethylated genes detected by DNA microarray and bisulfite pyrosequencing of bladder cancers and applied them ...

Urine and bladder washing cytology for detection of urothelial carcinoma: standard test with new possibilities

BACKGROUND Light microscopic evaluation of cell morphology in preparations from urine or bladder washing containing exfoliated cells is a standard and primary method for the detection of bladder cancer and also malignancy from other parts of the urinary tract. The cytopathologic examination is a valuable method to detect an early recurrence of malignancy or new primary carcinoma during the foll...

Carcinoembryonic antigen in the urine of patients with urothelial carcinoma.

Increased amounts of carcinoembryonic antigen (C.E.A.) or C.E.A.-like material are found in the urine of many patients with transitional cell carcinomas of the bladder, including those presumed to be at an early stage of development. It is suggested that measurement of urinary C.E.A. is of clinical diagnostic value in the detection and follow-up of urothelial carcinomas.

Urinary NID2 and TWIST1 methylation to augment conventional urine cytology for the detection of bladder cancer.

BACKGROUND Abnormal methylation of urinary TWIST1 and NID2 conferred high sensitivity and specificity for the detection of urothelial carcinoma. OBJECTIVE We examine the performance of the urine-based TWIST1/NID2 methylation assay with the addition of urine cytology for the detection of urothelial carcinoma. MATERIALS AND METHODS A prospective multi-institutional study was conducted to asse...
