Novel mumps virus epitopes reveal robust cytotoxic T cell responses after natural infection but not after vaccination

چکیده
Abstract Mumps is nowadays re-emerging despite vaccination. The contribution of T cell immunity to protection against mumps has not been clearly defined. Previously, we described a set 41 peptides that were eluted from human leukocyte antigen (HLA) class I molecules virus (MuV)-infected cells. Here, confirmed immunogenicity five novel HLA-B*07:02- and HLA-A*01:01-restricted MuV epitopes this peptides. High frequencies cells these could be detected ex vivo in all tested patients. Moreover, epitope-specific derived patients displayed strong cytotoxic activity. In contrast, only marginal responses recently vaccinated persons, corroborating earlier findings. Identifying which are dominantly targeted the mumps-specific CD8 + T- response an important step towards better understanding discrepancies between natural infection or vaccination-induced cell-mediated immune protection.

Human CD4+ T-cell repertoire of responses to influenza A virus hemagglutinin after recent natural infection.
The human CD4+ T-cell repertoire of responses to hemagglutinin (HA) of influenza virus A/Beijing/32/92 was examined 3 to 6 months after natural infection by using a panel of 16-mer peptides overlapping by 11 residues. Short-term CD4+ T-cell lines were derived by using full-length HAs of virus A/Beijing/32/92 from 12 unrelated, major histocompatibility complex (MHC) class I and II haplotyped adu...
متن کاملCD4+ T cells are not required for the induction of dengue virus-specific CD8+ T cell or antibody responses but contribute to protection after vaccination.
The contribution of T cells to the host response to dengue virus (DENV) infection is not well understood. We previously demonstrated a protective role for CD8(+) T cells during primary DENV infection using a mouse-passaged DENV strain and IFN-α/βR(-/-) C57BL/6 mice, which are susceptible to DENV infection. In this study, we examine the role of CD4(+) T cells during primary DENV infection. Four ...
متن کاملHuman CD4+ T Helper Cell Responses after Tick-Borne Encephalitis Vaccination and Infection
Tick-borne encephalitis virus (TBEV) is a human-pathogenic flavivirus that is endemic in large parts of Europe and Asia and causes severe neuroinvasive illness. A formalin-inactivated vaccine induces strong neutralizing antibody responses and confers protection from TBE disease. CD4+ T cell responses are essential for neutralizing antibody production, but data on the functionalities of TBEV-spe...
متن کاملBoosted Influenza-Specific T Cell Responses after H5N1 Pandemic Live Attenuated Influenza Virus Vaccination
BACKGROUND In a phase I clinical trial, a H5N1 pandemic live attenuated influenza virus (pLAIV) VN2004 vaccine bearing avian influenza H5N1 hemagglutinin (HA) and NA genes on the A/Ann Arbor cold-adapted vaccine backbone displayed very restricted replication. We evaluated T cell responses to H5N1 pLAIV vaccination and assessed pre-existing T cell responses to determine whether they were associa...
