چکیده
The dietary triacylglycerol (TAG) gets absorbed and accumulated in the body through monoacylglycerol (MAG) pathway, which plays a major role obesity related disorders. main enzyme of this acyltransferase 2 (MGAT2), is considered as potential target for developing antiobesity compounds. Hence, there need vitro cell-based assays screening leads MGAT2 inhibitors. Because synthetic inhibitor’s side effects, an increased interest natural extracts leads. we have optimized 2-MAG-induced TAG accumulation inhibitory assay to screen using HIEC-6 cell line. A concentration-dependent was observed when cells were fed with exogenous 2-MAG. confirmed by situ BODIPY staining quantified. However, no seen DAG or TAG, suggesting MGAT2-mediated MAG uptake its conversion TAG. We demonstrated utility five different plant-based aqueous extracts. These showed various inhibition levels (25% 30%) HIEC-6. these assay. This adds new methodology screening, developing, evaluating inhibitors addressing

Discovery of novel ansamycins possessing potent inhibitory activity in a cell-based oncostatin M signalling assay.
We describe the isolation and characterisation of novel non-benzoquinone ansamycin metabolites related to geldanamycin from a culture of Streptomyces sp. S6699. The compounds possess potent inhibitory activity in a cell-based assay measuring inhibition of oncostatin M signalling in a reporter cell line utilising a secreted placental alkaline phosphatase (sPAP) readout. In this paper we report t...
متن کاملA novel assay system for macrophage-activating factor activity using a human U937 cell line.
BACKGROUND Macrophages play important roles in antitumor immunity, and immunotherapy with the group-specific component protein-derived macrophage-activating factor (GcMAF) has been reported to be effective in patients with various types of cancers. However, in macrophage research, it is important to properly evaluate macrophage activity. MATERIALS AND METHODS U937 macrophages were induced by ...

In the recent decade, liver diseases with high mortality rate have became one of the major health problems worldwide. Liver transplantation is the most effective and the standard treatment for decompensated liver disease, but, shortage of available organs and inaccuracy in modeling of the liver diseases are the most limiting issues in treatment. Regenerative medicine and cell based therapy str...
متن کاملFunctionomics of NCC mutations in Gitelman syndrome using a novel mammalian cell-based activity assay.
Gitelman syndrome (GS) is an autosomal recessive salt-wasting tubular disorder resulting from loss-of-function mutations in the thiazide-sensitive NaCl cotransporter (NCC). Functional analysis of these mutations has been limited to the use of Xenopus laevis oocytes. The aim of the present study was, therefore, to analyze the functional consequences of NCC mutations in a mammalian cell-based ass...
متن کاملCell-based Models To Investigate Tau Aggregation
Accumulation of abnormal tau aggregates in neuron is an important pathological signature in multiple neurodegenerative disorders including Alzheimer's disease. Tau is a neuron specific microtubule-associated protein that regulates microtubule stability, which is critical for axonal outgrowth and synaptic plasticity. In a pathological condition, tau dissociates from microtubules and forms insolu...
