Non‐specific phospholipases C2 and C6 redundantly function in pollen tube growth via triacylglycerol production in Arabidopsis

چکیده
Non-specific phospholipase Cs (NPCs) are responsible for membrane lipid remodeling that involves hydrolysis of the polar head group phospholipids. Arabidopsis NPC2 and NPC6 essential in gametogenesis, but their underlying role remains elusive. Here, we show these NPCs required triacylglycerol (TAG) production pollen tube growth. highly expressed developing tubes localized at endoplasmic reticulum. Mutants showed reduced rate germination, length amount droplets (LDs). Overexpression or induced LD accumulation, which suggests involved production. Furthermore, mutants defective biosynthesis TAG, a major component LDs, These results suggest gametogenesis hydrolyzing phospholipids producing TAG Thus, from to during growth represents an emerging NPC family plant developmental control.

Five gametophytic mutations affecting pollen development and pollen tube growth in Arabidopsis thaliana.
Mutant analysis represents one of the most reliable approaches to identifying genes involved in plant development. The screening of the Versailles collection of Arabidopsis thaliana T-DNA insertion transformants has allowed us to isolate different mutations affecting male gametophytic functions and viability. Among several mutated lines, five have been extensively studied at the genetic, molecu...
متن کاملGibberellin regulates pollen viability and pollen tube growth in rice.
Gibberellins (GAs) play many biological roles in higher plants. We collected and performed genetic analysis on rice (Oryza sativa) GA-related mutants, including GA-deficient and GA-insensitive mutants. Genetic analysis of the mutants revealed that rice GA-deficient mutations are not transmitted as Mendelian traits to the next generation following self-pollination of F1 heterozygous plants, alth...
متن کاملThe Arabidopsis GPR1 Gene Negatively Affects Pollen Germination, Pollen Tube Growth, and Gametophyte Senescence
Genes essential for gametophyte development and fertilization have been identified and studied in detail; however, genes that fine-tune these processes are largely unknown. Here, we characterized an unknown Arabidopsis gene, GTP-BINDING PROTEIN RELATED1 (GPR1). GPR1 is specifically expressed in ovule, pollen, and pollen tube. Enhanced green fluorescent protein-tagged GPR1 localizes to both nucl...
متن کاملArabidopsis FIMBRIN5, an actin bundling factor, is required for pollen germination and pollen tube growth.
Actin cables in pollen tubes serve as molecular tracks for cytoplasmic streaming and organelle movement and are formed by actin bundling factors like villins and fimbrins. However, the precise mechanisms by which actin cables are generated and maintained remain largely unknown. Fimbrins comprise a family of five members in Arabidopsis thaliana. Here, we characterized a fimbrin isoform, Arabidop...
متن کاملIons and Pollen Tube Growth
Ions play a crucial role in the control of pollen tube growth. In this review we focus on four that seem especially important: calcium (Ca2+), protons (H+), potassium (K+), and chloride (Cl–). Ca2+ in the extracellular medium is essential for growth; it forms a steep intracellular tip-focused gradient, and exhibits a prominent extracellular tip-directed Ca2+ influx. pH is also essential for gro...
