Nicotinamide phosphoribosyltransferase inhibitor is a potential therapeutic target in LPS-induced human trophoblast cell injury

Purpose: To investigate the role of nicotinamide phosphoribosyltransferase (NAMPT) in lipopolysaccharide (LPS)-induced damage trophoblastic HTR-8/SVneo cells (HTR8 cells), with aim ultimately providing new therapeutic targets preeclampsia (PE).Methods: Trophoblastic was cultured and treated LPS to mimic PE vitro, while FK866, an antagonist NAMPT, used establish inflammatory model HTR8 cells. Western blot, enzyme-linked immunosorbent assay (ELISA) quantitative real-time-polymerase chain reaction (qRT-PCR) were evaluate response cells, cell counting kit-8 (CCK8) oxidative stress kits performed quantify activity HTR cells.Results: Compared control group, administration significantly increased expression levels NAMPT FK866 suppressed proinflammatory factors IL-1β, TNF-α IL-6, alleviated inflammation by inhibiting NAMPT-mediated NF-κB pathway. The antioxidant effect achieved via activation proteins,catalase (CAT) glutathione (GSH).Conclusion: protects from LPS-induced through inhibition NAMPT-NF-κB signaling Thus, is a potential target for (PE).
