Mycobiota Associated with Symptomatic and Asymptomatic Fraxinus excelsior in Post-Dieback Forest Stands

In Lithuania, the dieback of European ash (Fraxinus excelsior L.), caused by alien ascomycete Hymenoscyphus fraxineus, started in mid-1990s, resulting a large-scale decline F. and its dominated forest habitats. Nevertheless, recent inventories show presence several hundred hectares naturally regenerated stands. We used seven sites three planted progeny trials to collect leaves, shoots, roots, surrounding soil study ash-associated fungal communities based on high-throughput sequencing. Results showed that associated with re-emerging stands post-dieback areas were composed 1487 taxa. Among these, 60.5% Ascomycota, 37.5%—Basidiomycota, 1.7%—Zygomycota, 0.2% Chytridiomycota. Revealed mycobiota was largely endophytic as these Cladosporium sp., Fraxinicola fraxini (syn. Venturia fraxini) Vishniacozyma foliicola. Identified also included range ash-specific fraxineus occurred all but not frequent. sp. strongest negative correlation H. fraxineus. This ascomycete, given dominance shoots organic layer, might be limiting factor for infection rate or spread Although asymptomatic symptomatic samples differed significantly from each other, majority most frequently found taxa host-specific, suggesting negligibly affected dieback. Investigated natural habitats exhibited larger diversity (especially ash-specific), than former grasslands, indicating importance restoration programs.
