چکیده

Abstract Background Receptive endometrium is a restraining factor in the establishment of pregnancy several estrogen-dependent gynecological disorders including uterine leiomyomas. Recently, data are beginning to accrue suggesting negative impact non-cavity distorting intramural fibroids on molecular mediators endometrial receptivity. The potential importance mammalian target rapamycin (mTOR) signaling pathway has been suggested during embryo implantation. However, its exact role fibroid-associated window implantation poorly defined. objective study was examine expression and cellular distribution key components mTOR infertile women with leiomyomas ( n = 24) as compared fertile controls 17). Relative gene analysis , TSC1 TSC2 performed by real-time PCR. Immunohistochemistry used evaluate mTOR, phospho-mTOR (Serine 2448), TSC1, TSC2, phospho-TSC2 (Threonine 1462), phospho-S6 ribosomal protein (Serines 235 236) Ki67. Results In comparison controls, statistically significant upregulation (8.97-fold; p < 0.001) downregulation mRNA (− 6.01-fold; 0.01) levels cell-specific proteins phospho-mTOR, phospho-TSC2, were observed women. ratio p-mTOR/mTOR p-TSC2/TSC2 significantly higher Pearson’s correlation revealed between p-mTOR positive p-S6 group. Increased Ki67 labelling index glandular epithelium (GE) stroma from controls. Conclusions Loss function enhanced activated downstream targets, group, indicate heightened which might contribute impaired number Ki67-positive nuclei suggests that may help drive dysregulated proliferation midsecretory leading compromised fertility present findings provide avenues for future investigation nonsurgical alternative treatment infertility these patients.

assessment of endometrial-subendometrial blood flow detected by color doppler sonography and uterine receptivity in infertile women

ndometrial vasculature has been shown to play an important role in the early endometrial response to the implanting blastocyst, and vascular changes may contribute to uterine receptivity . the aim of this study was to investigate the role of endometrial and subendometrial vascularity and thickness in uterine receptivity in infertile women treated with assisted reproductive technologies (art). a...

PI3K/Akt/mTOR signaling & its regulator tumour suppressor genes PTEN & LKB1 in human uterine leiomyomas

BACKGROUND & OBJECTIVES Despite their high occurrence and associated significant level of morbidity manifesting as spectrum of clinical symptoms, the pathogenesis of uterine leiomyomas (ULs) remains unclear. We investigated expression profile of tumour suppressor genes PTEN (phosphatase and tensin homolog deleted on chromosome ten) and LKB1 (liver kinase B1), and key signaling components of P13...

Endometrial Histological Findings in Infertile Women

Background and Objective: Infertility is a global health problem and in Pakistan frequency of subfertility is 21.9%. The fact that cyclical histological changes in the endometrium correlate with fluctuating biorhythms of various hormone levels enables histological appearance of the endometrium to be a reliable parameter to evaluate the cause of infertility. Objective: To discern the histologica...

Endometrial markers of uterine receptivity utilizing the donor oocyte model.

BACKGROUND Ethical constraints limit the ability to study peri-implantation phase human endometrium. In this study, the donor oocyte model was used to study candidate endometrial markers of uterine receptivity. METHODS Archived, paraffin-embedded tissue obtained by endometrial biopsy during cycle days 21-23 of patients undergoing 'mock' hormonal treatment cycles were evaluated by standard his...

Improved Fertility Following Enucleation of Intramural Myomas in Infertile Women

BACKGROUND The relationship between intramural myomas and fertility remains unclear. The main debate rests on whether cavity-distorting intramural myomas (CDMs) adversely affect fertility more than non-CDMs. We aimed to compare the effects of enucleating non-CDMs and CDMs on fertility improvement in females with unexplained infertility. METHODS We prospectively recruited 83 women undergoing m...
