چکیده
Abstract Lowered glucose availability, sensed by the hindbrain, has been suggested to enhance gluconeogenesis and food intake as well suppress reproductive function. In fact, our previous histological in vitro studies suggest that hindbrain ependymal cells function a sensor. The present study aimed clarify sensor-hypothalamic neural pathway activated response glucoprivation mediate counterregulatory physiological responses. Administration of 2-deoxy-D-glucose (2DG), an inhibitor utilization, into fourth ventricle (4V) male rats for 0.5 hour induced messenger RNA (mRNA) expression c-fos, marker cellular activation, 4V, but not lateral ventricle, third or central canal without significant change blood testosterone levels. 2DG 4V 1 significantly increased levels, intake, decreased Simultaneously, c-Fos protein was detected cells; dopamine ?-hydroxylase-immunoreactive C1, C2, A6 regions; neuropeptide Y (NPY) mRNA-positive C2; corticotropin-releasing hormone (CRH) hypothalamic paraventricular nucleus (PVN); NPY arcuate (ARC). Taken together, these results lowered cells, activates catecholaminergic and/or neurons followed CRH PVN ARC, thereby leading responses, such enhancement gluconeogenesis, suppression sex steroid secretion.

Localized glucoprivation of hindbrain sites elicits corticosterone and glucagon secretion.
Glucose is required for brain energy metabolism. Decerebration, aqueduct occlusion, and cannula mapping studies have established that glucose-sensing cells capable of eliciting feeding and adrenal medullary responses to glucoprivation are localized in the hindbrain. Glucoprivation also evokes corticosterone and glucagon secretion, but the location of receptors mediating these responses is unkno...
متن کاملAntecedent hindbrain glucoprivation does not impair the counterregulatory response to hypoglycemia.
Recurrent hypoglycemia impairs hormonal counterregulatory responses (CRRs) to further bouts of hypoglycemia. The hypothalamus and hindbrain are both critical for sensing hypoglycemia and triggering CRRs. Hypothalamic glucose sensing sites are implicated in the pathogenesis of defective CRRs; however, the contribution of hindbrain glucose sensing has not been elucidated. Using a rat model, we co...
متن کاملHypothalamic Nutrient Sensing Activates a Forebrain-Hindbrain Neuronal Circuit to Regulate Glucose Production In Vivo
OBJECTIVE Hypothalamic nutrient sensing regulates glucose production, but the neuronal circuits involved remain largely unknown. Recent studies underscore the importance of N-methyl-d-aspartate (NMDA) receptors in the dorsal vagal complex in glucose regulation. These studies raise the possibility that hypothalamic nutrient sensing activates a forebrain-hindbrain NMDA-dependent circuit to regula...

THE CENTRAL NERVOUS SYSTEM detects correlates of reduced energy availability and activates circuits that trigger compensatory responses that collectively restore energy balance. The seminal experiments of Claude Bernard and Walter Cannon defined concepts underlying the regulatory physiology of energy balance, an area that continues to attract investigators interested in addressing a range of im...
متن کاملParticipation of Hindbrain AMP-Activated Protein Kinase in Glucoprivic Feeding
OBJECTIVE To examine the role of AMP-activated protein kinase (AMPK) in the control of glucoprivic feeding by hindbrain catecholamine neurons. RESEARCH DESIGN AND METHODS Micropunched hindbrain samples were collected from control and 2-deoxy-d-glucose (2DG)-injected rats for Western blot analysis of phosphorylated (activated) AMPK (pAMPK). Samples also were collected from 2DG-injected rats pr...
