Moleküler markörler kullanarak çerezlik kabaklarda (Cucurbita pepo L.) safl?k düzeylerinin tahmin edilmesi

Amaç: Bu çal??mada moleküler markörler kullan?larak çerezlik kabaklarda (Cucurbita pepo L.) safl?k düzeylerinin tahmin edilmesi amaçlanm??t?r.Yöntem ve Bulgular: Dominant kodominant markör teknikleri (simple sequence repeats, SSR inter-simple ISSR) yard?m?yla S0 (hiç kendileme yap?lmam??), S1, S2 S3 kademesindeki kabaklar?n düzeyleri ile heterozigotluk aras?ndaki ili?ki edilmi?tir. analizleri sonucunda CMTp182 CMTm66 primerlerinde ortalama PIC de?eri (0.9) gen çe?itlili?i (0.10) belirlenmi?tir. ISSR HVH(CA)7T, HVH(TCC)7 BDB(CA)7C en yüksek (0.4) bulunmu? olup s?ras?yla 0.61, 0.67 0.86 olarak tespit kullan?lan verimlili?inin bir ifadesi olan de?erleri kar??la?t?r?ld???nda primerlerinin 0.57 bulunurken, markörlerinin ise 0.2 hesaplanm??t?r. Safla?ma çal??malar?nda polimorfizm göstermeleri nedeniyle daha etkin oldu?u saptanm??t?r.Genel Yorum: Yap?lan analizler farkl? kademeler (S0, S3) aras?nda heterozigot bant say?s? giderek azalan e?ilim göstermi?tir. En fazla kademesinden az elde durum safla?ma oran? artt?kça say?s?n?n azald???n? göstermektedir. Ba?ar?l? bulunulan primerlerin düzeyi belirleme kullan?labilece?i sonucuna var?lm??t?r.Çal??man?n Önemi Etkisi: Bitkilerde saf hatlar?n ?slah çal??malar? için çok önemli etkendir. Çerezlik kabakta hibrit tohum üretiminde ebeveynlerin kullan?lmas? önemlidir bitkilerde düzeylerini etmenin pratik yolu bulunmamaktad?r. Arazide tohumlardan edilen bitkiler morfolojik özellikleri dikkate alarak genetik aç?l?m olmad??? anla??labilmektedir. Moleküler markörlerin belirlenmesinde kullan?labilirli?inin belirlenmi? olmas? önemlidir.
