Molecular profiling of bone remodeling occurring in musculoskeletal tumors

Musculoskeletal malignancy is often accompanied by aberrant bone remodeling, leading to tumor cell invasion into skeletal tissues and causing severe pain. BMPs, FGF-2, RANKL have been identified as promising regulators in physiological remodeling. In this study, we explored the expressional profile of 1361 patients with 22 varieties musculoskeletal tumors. Notably, expression FGF-2 was under detected all patients. Among BMP1 BMP15, found that BMP1, BMP2, BMP4, BMP5, BMP6, BMP7 were prevalent. comparison normal bones, osteosarcoma highly expressed statistical significance. Synovial sarcoma upregulated BMP7; rhabdomyosarcoma increased BMP4; alveolar soft part BMP7. To visualize BMP-oriented interactions a microenvironment, developed novel software analyzes numerous cell-to-cell ligand-to-receptor interactions, is, Environmentome, delineating osteosarcoma-secreted BMP-4 synovial sarcoma-secreted potently interact osteoblasts, osteocytes, osteoclast precursors, mature osteoclasts. Specifically, quantification analysis revealed relationship between osteoclast/precursor, BMP4–BMPR2 BMP4–ACVR2A most potent. Regarding association osteocyte/osteoblast, BMP4–ACVR1 key interactions. connection BMP7–ACVR2A BMP7–BMPR2 remarkable. With regard cellular link potent interaction. conclusion, our new outlook suggests delivering pathological events clinically underlie behind pain or fracture
