چکیده

Abstract Knockdown resistance (kdr) is a common mechanism of insecticide in head lice to the conventionally used pyrethroid pediculosis and can be result various amino acid substitutions within voltage-sensitive sodium channel (VSSC). In this study, 54 sequences from varied specimens were investigated monitor well-known mutations probable new mutations. The Pediculus humanus capitis de Geer collected 13 provinces Iran. stored 70% ethanol until DNA extraction PCR amplification ~900-bp fragment VSSC. analyzed using different bioinformatics software for detection kdr additional potentially associated with lice. There six an old (haplotype I) haplotypes Iranian louse population. K794E, F815I, N818D reported first time. P813H mutation was most prevalent substitution eight provinces. Among 53 sequences, 26 (49%) homozygous susceptible, 27 (51%) heterozygotes. Thus, 51% Iran harbored only allele. exact test Hardy–Weinberg (H–W) equilibrium showed that genotype frequencies differed significantly expectation East-Azerbaijan Tehran Moreover, these populations had inbreeding coefficient (Fis) <0, indicating excess This observation suggests are currently under active selective pressure. For rest populations, H–W expectations harmony. results current study highlight molecular techniques accurate genotypes before their establishment Accurate resistant seems helpful decision-making on control programs monitoring management.

Knockdown resistance allele frequencies in North American head louse (Anoplura: Pediculidae) populations.

The study examines the extent and frequency of a knockdown-type resistance allele (kdr type) in North American populations of human head lice. Lice were collected from 32 locations in Canada and the United States. DNA was extracted from individual lice and used to determine their zygosity using the serial invasive signal amplification technique to detect the kdr-type T917I (TI) mutation, which ...

High diversity and rapid diversification in the head louse, Pediculus humanus (Pediculidae: Phthiraptera)

The study analyzes sequence variation of two mitochondrial genes (COI, cytb) in Pediculus humanus from three countries (Egypt, Pakistan, South Africa) that have received little prior attention, and integrates these results with prior data. Analysis indicates a maximum K2P distance of 10.3% among 960 COI sequences and 13.8% among 479 cytb sequences. Three analytical methods (BIN, PTP, ABGD) reve...

Expansion of the Knockdown Resistance Frequency Map for Human Head Lice (Phthiraptera: Pediculidae) in the United States Using Quantitative Sequencing

Pediculosis is a prevalent parasitic infestation of humans, which is increasing due, in part, to the selection of lice resistant to either the pyrethrins or pyrethroid insecticides by the knockdown resistance (kdr) mechanism. To determine the extent and magnitude of the kdr-type mutations responsible for this resistance, lice were collected from 138 collection sites in 48 U.S. states from 22 Ju...

Permethrin Resistance Due to Knockdown Gene Mutations is Prevalent in Human Head Louse Populations

Permethrin resistance in head lice is mainly conferred by the knockdown resistance (kdr) trait, a voltagesensitive sodium channel (VSSC) insensitivity factor. Three VSSC mutations have been identified and confirmed to reduce the sensitivity of VSSC to permethrin. A step-wise resistance monitoring system has been established based on molecular resistance detection techniques. Quantitative sequen...

Geographical distribution of pyrethroid resistance allele frequency in head lice (Phthiraptera: Pediculidae) from Argentina.

The human head louse, Pediculus humanus capitis De Geer (Phthiraptera: Pediculidae), is an obligate ectoparasite that causes pediculosis capitis and has parasitized humans since the beginning of humankind. Head louse infestations are widespread throughout the world and have been increasing since the early 1990s partially because of ineffective pediculicides. In Argentina, the overuse of product...
