Molecular Insights into the Flavivirus Replication Complex

چکیده

Flaviviruses are vector-borne RNA viruses, many of which clinically relevant human viral pathogens, such as dengue, Zika, Japanese encephalitis, West Nile and yellow fever viruses. Millions people infected with these viruses around the world each year. Vaccines only available for some members this large virus family, there no effective antiviral drugs to treat flavivirus infections. The unmet need vaccines therapies against flaviviral infections drives research towards a better understanding epidemiology, biology immunology flaviviruses. In review, we discuss basic replication process focus on molecular aspects genome replication. Within virus-induced intracellular membranous compartments, takes place, starting from poly protein expression processing assembly complex, followed by delivery progeny particle sites. We attempt update latest key events during highlight knowledge gaps future studies.

Flavivirus replication complex assembly revealed by DNAJC14 functional mapping.

DNAJC14 is an Hsp40 family member that broadly modulates flavivirus replication. The mechanism by which DNAJC14 stoichiometrically participates in flavivirus replication complex (RC) formation is unknown; both reduced and elevated levels result in replication inhibition. Using yellow fever virus (YFV), we demonstrate that DNAJC14 redistributes and clusters with YFV nonstructural proteins via a ...

Structural analysis of the human SYCE2–TEX12 complex provides molecular insights into synaptonemal complex assembly

The successful completion of meiosis is essential for all sexually reproducing organisms. The synaptonemal complex (SC) is a large proteinaceous structure that holds together homologous chromosomes during meiosis, providing the structural framework for meiotic recombination and crossover formation. Errors in SC formation are associated with infertility, recurrent miscarriage and aneuploidy. The...

Structural insights into eukaryotic DNA replication

THREE DNA POLYMERASES OF THE B FAMILY FUNCTION AT THE REPLICATION FORK IN EUKARYOTIC CELLS: DNA polymerases α, δ, and ε. DNA polymerase α, an heterotetramer composed of two primase subunits and two polymerase subunits, initiates replication. DNA polymerases δ and ε elongate the primers generated by pol α. The DNA polymerase from bacteriophage RB69 has served as a model for eukaryotic B family p...

Newer Insights into the Prsevention of the Common Cold

Since most colds are self-limiting, symptoms will usually resolve on their own in seven to ten days. For the majority of patients, treatment is symptomatic and targeted at the most bothersome symptoms. Specific nonprescription therapy may relieve cold symptoms but should be used with caution, especially in at-risk patient populations. Patients should monitor for worsening symptoms, and progress...

Crystal structure of the GINS complex and functional insights into its role in DNA replication.

The GINS complex, which contains the four subunits Sld5, Psf1, Psf2, and Psf3, is essential for both the initiation and progression of DNA replication in eukaryotes. GINS associates with the MCM2-7 complex and Cdc45 to activate the eukaryotic minichromosome maintenance helicase. It also appears to interact with and stimulate the polymerase activities of DNA polymerase epsilon and the DNA polyme...
