چکیده

Polycystic ovary syndrome (PCOS) is typically characterized by a polycystic ovarian morphology, hyperandrogenism, ovulatory dysfunction, and infertility. Furthermore, PCOS patients undergoing stimulation have more oocytes; however, the poor quality of oocytes leads to lower fertilization implantation rates, decreased pregnancy increased miscarriage rates. The complex molecular mechanisms underlying remain be elucidated. We obtained matched cumulus cells (CCs) from patients, compared them with age-matched controls, performed RNA sequencing analysis explore transcriptional characteristics their CCs. Moreover, we validated our newly confirmed candidate genes for immunofluorescence. Unsupervised clustering showed that overall global gene expression patterns transposable element (TE) profiles tightly clustered together, clearly distinct those controls. Abnormalities in functionally important pathways are found oocytes. Notably, involved microtubule processes, TUBB8 TUBA1C , overexpressed metabolic oxidative phosphorylation also dysregulated both CCs patients. oocytes, differentially expressed TEs not uniformly dispersed human chromosomes. Endogenous retrovirus 1 ( ERV1 ) elements located on chromosomes 2, 3, 4, 5 rather highly upregulated. Interestingly, these correlate most protein-coding genes, including tubulin-associated TUBB8P8 linking occurrence PCOS. Our comprehensive CCs, TE expression, revealed specific features aberrantly elevated provides additional markers may contribute compromised oocyte developmental competence findings implications treatment strategies improve maturation outcomes women

Molecular abnormalities in oocytes from women with polycystic ovary syndrome revealed by microarray analysis.

CONTEXT Polycystic ovary syndrome (PCOS), the most common cause of anovulatory infertility, is characterized by increased ovarian androgen production and arrested follicle development and is frequently associated with insulin resistance. These PCOS phenotypes are associated with exaggerated ovarian responsiveness to FSH and increased pregnancy loss. OBJECTIVE The objective of this study was t...

O-21: Differential Expression and Epigenetic Pattern of HOX Family Genes in Cumulus Cells of Mature MII Oocytes from Patients with Polycystic Ovary Syndrome

Background Ovarian tissue cryopreservation represents a promising strategy to preserve the ovarian function in cancer patients. It is usually performed by slow freezing/rapid thawing (SF/RT). Recent studies emphasize an ultrarapid cryopreservation procedure, vitrification/warming (V/W), since it might prevent damages due to ice crystal formation. Comparative studies between the cryopreservation...

O-29: Differences in The Transcriptional Profiles of Human Cumulus Cells Isolated From MI and MII Oocytes of Patients with Polycystic Ovary Syndrome

Background: Polycystic ovary syndrome (PCOS) is a common endocrine and metabolic disorder in women. The abnormalities of endocrine and intra-ovarian paracrine interactions may change the microenvironment for oocyte development during the folliculogenesis process and reduce the developmental competence of oocytes in PCOS patients who are suffering from anovulatory infertility and pregnancy loss....

Gene expression microarray profiles of cumulus cells in lean and overweight-obese polycystic ovary syndrome patients.

The aim of this work was to study gene expression patterns of cultured cumulus cells from lean and overweight-obese polycystic ovary syndrome (PCOS) patients using genome-wide oligonucleotide microarray. The study included 25 patients undergoing in vitro fertilization and intra-cytoplasmic sperm injection: 12 diagnosed with PCOS and 13 matching controls. Each of the groups was subdivided into l...
