Molecular Detection of Extended Spectrum Beta-Lactamase Gene in Clinical Isolates of Klebsiella Species in Jos North Central of Nigeria

Aims: The aim of the study was to determine antimicrobial susceptibility Klebsiella species, prevalence Extended Spectrum Beta-Lactamases (ESBLs) and gene coding ESBL production in Species obtained from clinical specimens patients attending Plateau State Specialist Jos University Teaching Hospitals both North Central Nigeria.
 Study Design: This hospital based conducted Department Medical Microbiology unit Hospital (JUTH) during periods January April 2020.
 Methodology: Out 156 non-motile, oxidase negative gram organisms collected, 137 isolates were identified be species using oxoid Microbact 24E kit for Enterobacteriaceae identification Microsoft bacteria identification. Antimicrobial Susceptibility Testing (AST) screening done by Kirby-Bauer disk diffusion method Single antibiotics which include Ceftazidime (30µg), Cefotaxime Imipenem (10µg), Ciprofloxacin (5µg), Gentamicin Ampicillin (10 µg), Aztreonam (30 Azithromycin (15 µg) on Mueller Hinton Agar (oxoid).Break point determined CLSI AST guidelines. that showed resistance or reduced are suspected positive Klebsiella. Phenotypic confirmation carried out Double Disk Synergy Test (DDST) with 30µg (oxoid), (oxoid) Amoxicillin Clavulanic acid 20/10ug (Oxoid).Genotypic detection Polymerase Chain Reaction (PCR).
 Results: highly sensitive pneumonia 100% sensitivity. Gentamicin, Ampicillin, susceptibility. Genotypic results presence bla TEM, SHV genes at 868bp 972bp band size no occurrence CTX-M gene. There multiple some tested while recorded 15.3% this study.
 Conclusions: is low compared other studies states within country. Efforts should put place ensure non-proliferation strain as it creates not only a therapeutic challenge but health problem.
