چکیده
Introduction Vacuolar protein sorting 29 (VPS29) is a core component of the retromer-retriever complex and essential for recycling numerous cell-surface cargoes from endosomes. However, there are no reports yet on VPS29 Eimeria spp. Methods Here, we cloned prokaryotically expressed partial sequence tenella (EtVPS29) with RT-PCR engineered strain Escherichia coli respectively. The localization in E. sporozoites was investigated immunofluorescence (IFA) overexpression assays. And its protective efficacy against infection chickens animal protection test. Results An EtVPS29 gene fragment an ORF reading frame 549 bp cloned. band size recombinant protein, rEtVPS29, approximately 39 kDa recognized by chicken anti- positive serum. observed widely distributing cytoplasm IFA rEtVPS29 significantly increased average body weight gain decreased mean lesion score oocyst output chickens. relative rate rEtVPS29-immunized group 62.9%, which higher than that unimmunized challenged (P < 0.05). percentage reduced immunized 32.2%. anticoccidial index 144.2. Molecular Characterization and Functional Analysis of a Novel Calcium-Dependent Protein Kinase 4 from Eimeria tenella
Eimeria tenella is an obligate intracellular parasite that actively invades cecal epithelial cells of chickens. The basis of cell invasion is not completely understood, but some key molecules of host cell invasion have been discovered. This paper investigated the characteristics of calcium-dependent protein kinase 4 (EtCDPK4), a critical molecule in E. tenella invasion of host cells. A full-len...
متن کاملMolecular Characterization and Analysis of a Novel Protein Disulfide Isomerase-Like Protein of Eimeria tenella
Protein disulfide isomerase (PDI) and PDI-like proteins are members of the thioredoxin superfamily. They contain thioredoxin-like domains and catalyze the physiological oxidation, reduction and isomerization of protein disulfide bonds, which are involved in cell function and development in prokaryotes and eukaryotes. In this study, EtPDIL, a novel PDI-like gene of Eimeria tenella, was cloned us...
متن کاملPartial purification and characterization of an aminopeptidase from Eimeria tenella.
Our previous investigation demonstrated the expression in Eimeria tenella sporulated oocysts of an aminopeptidase (AP) with strong homology to AP N. To further understand the role of proteases during development, we investigated the molecular and biochemical properties of E. tenella AP. Greater than 95% AP activity was present in a soluble extract during sporulation of oocysts with highest acti...
متن کاملMolecular Characterization and Immune Protection of a New Conserved Hypothetical Protein of Eimeria tenella
The genome sequences of Eimeria tenella have been sequenced, but >70% of these genes are currently categorized as having an unknown function or annotated as conserved hypothetical proteins, and few of them have been studied. In the present study, a conserved hypothetical protein gene of E. tenella, designated EtCHP559, was cloned using rapid amplification of cDNA 5'-ends (5'RACE) based on the e...
متن کاملTrafficking of vacuolar proteins: the crucial role of Arabidopsis vacuolar protein sorting 29 in recycling vacuolar sorting receptor.
The retromer is involved in recycling lysosomal sorting receptors in mammals. A component of the retromer complex in Arabidopsis thaliana, vacuolar protein sorting 29 (VPS29), plays a crucial role in trafficking storage proteins to protein storage vacuoles. However, it is not known whether or how vacuolar sorting receptors (VSRs) are recycled from the prevacuolar compartment (PVC) to the trans-...
