Molecular characterization and pathogenicity of an infectious cDNA clone of tomato brown rugose fruit virus

Abstract Tomato brown rugose fruit virus (ToBRFV) is a new member of the genus Tobamovirus , and has potential to affect production marketability tomatoes peppers. In this study, we sequenced analyzed complete genome ToBRFV isolates from tomato plants showing mosaic mottling symptoms in Yunnan Province China. We constructed full-length infectious cDNA clone ToBRFV, which could induce systemic infection with typical tomato, Nicotiana benthamiana N. tabacum cv. Samsun nn through Agrobacterium -mediated inoculation. Further experimental evidence demonstrated that rod-shaped virions accumulating agroinfiltrated are sap-transmissible. This first report on construction biologically active, ToBRFV. The system developed herein will facilitate further research functions ToBRFV-encoded proteins plant-ToBRFV interactions reverse genetic approaches.
