چکیده
Abstract Background Intramuscular fat (IMF) content is a vital parameter for assessing pork quality. Increasing evidence has shown that microRNAs (miRNAs) play an important role in regulating porcine IMF deposition. Here, novel miRNA implicated adipogenesis was found, and its effect regulatory mechanism were further explored with respect to intramuscular preadipocyte proliferation differentiation. Results By adipose tissue sequencing analysis, we found miR-146a-5p potential regulator of adipogenesis. Further studies showed mimics inhibited differentiation, while the inhibitor promoted cell adipogenic Mechanistically, suppressed by directly targeting SMAD family member 4 ( SMAD4 ) attenuate TGF-β signaling. Moreover, differentiation preadipocytes TNF receptor-associated factor 6 TRAF6 weaken AKT/mTORC1 signaling downstream pathway. Conclusions MiR-146a-5p targets inhibit attenuating signaling, respectively. These findings provide biomarker promote

miR-425-5p Inhibits Differentiation and Proliferation in Porcine Intramuscular Preadipocytes
Intramuscular fat (IMF) content affects the tenderness, juiciness, and flavor of pork. An increasing number of studies are focusing on the functions of microRNAs (miRs) during porcine intramuscular preadipocyte development. Previous studies have proved that miR-425-5p was enriched in porcine skeletal muscles and played important roles in multiple physiological processes; however, its functions ...
متن کاملmiR-424-5p promotes proliferation of gastric cancer by targeting Smad3 through TGF-β signaling pathway
MiRNAs have been reported to regulate gene expression and be associated with cancer progression. Recently, miR-424-5p was reported to play important role in a variety of tumors. However, the role and molecular mechanisms of miR-424-5p in GC (gastric cancer) remains largely unknown. In this study, we aimed to explore the role of miR-424-5p in GC. QRT-PCR was used to determine the expression leve...
متن کاملmiR-429 Inhibits Differentiation and Promotes Proliferation in Porcine Preadipocytes
MicroRNAs (miRNAs) are crucial regulatory molecules for adipogenesis. They contribute to the controlling of proliferation and differentiation of preadipocytes. Previous studies revealed an important role of miR-429 in cell invasion, migration, and apoptosis. Our previous work has shown that the expression of miR-429 in subcutaneous fat can be observed in newly born (3-day-old) Rongchang piglets...
متن کاملMicroRNA-146a-5p Limits Elevated TGF-β Signal during Cell Senescence
MicroRNAs (miRNAs) are small non-coding RNAs implicated in post-transcriptional suppression of target genes in a sequencespecific manner and are thereby able to modulate cellular processes. miR-146a seems to play multiple roles in terms of regulating different phenotypes by targeting a wide range of different genes in various cellular contexts. Notably, miR-146a-5p targets Smad4 and tumor necro...
متن کاملMiR-146a-5p inhibits cell proliferation and cell cycle progression in NSCLC cell lines by targeting CCND1 and CCND2
Previous studies have indicated that miR-146a-5p acts as an oncogene in several types of cancer, yet a tumor suppressor gene in others. In non-small cell lung cancer (NSCLC), one report showed that it was downregulated and played the role of tumor suppressor. However, another study showed that miR-146a-5p was overexpressed in the serum of NSCLC patients compared to healthy controls. Therefore, ...
