Mineral oil interferes with potato virus Y in aphid stylets

چکیده
Abstract Mineral oils have been used for many decades to control the transmission of plant viruses and their ensuing epidemics. Although it is not clearly defined, mode action these has extensively studied, effects on feeding behaviour orientation vector or virus by its often put forward as possible explanations. This study examines effect mineral oil interaction between potato Y (PVY) vector, green peach aphid Myzus persicae , with particular focus organ. A lipophilic staining technique was visualize inside insect's stylets first time. Virus binding localization were also examined in following PVY acquisition. The results indicate that presence leaves reduces likelihood acquiring virus. verified this reduced acquisition could be accompanied a reduction viral load, measured reverse transcription‐quantitative PCR stylets. retention measured. mechanism which impacts seems take place very early stages acquisition, well before attaches

Aphid transmission studies using helper component proteins of potato virus Y expressed from a vector derived from potato virus X.
The genes encoding the helper component (HC) proteins of two strains of Potato virus Y (PVY) were cloned and the proteins expressed from a vector derived from Potato virus X (PVX). The expressed HC contained six N-terminal histidine residues to facilitate purification by metal affinity chromatography. Approximately 2-4 microg/g of purified HC was obtained from leaves of Nicotiana benthamiana pl...
متن کاملHeterologous Expression of Potato Virus Y Coat Protein, Isolate Pot187
Background: The advent of recombinant DNA technology has facilitated heterologous expression of proteins from various sources in different host systems including Escherichia coli. If a plant virus coat protein is expressed in the bacterium it can be used as the antigen for antibody preparation. Such a recombinant antigen preparation can be particularly useful where equipment such as ultracentri...
متن کاملSequence characteristics of potato virus Y recombinants.
Potato virus Y (PVY) is one of the most economically important plant pathogens. The PVY genome has a high degree of genetic variability and is also subject to recombination. New recombinants have been reported in many countries since the 1980s, but the origin of these recombinant strains and the physical and evolutionary mechanisms driving their emergence are not clear at the moment. The replic...

متن کاملA Novel Recombined Potato virus Y Isolate in China
This study reports the findings of a distinct Potato virus Y (PVY) isolate found in Northeast China. One hundred and ten samples (leaves and tubers) were collected from potato plants showing mosaic symptoms around the city of Harbin in Heilongjiang province of China. The collected tubers were planted and let to grow in a greenhouse. New potato plants generated from these tubers showed similar s...
